İneklerde kızgınlığın farklı devrelerinde elde edilen serviks ve vagina akıntılarında fiziksel ve kimyasal özelliklerle feromonların araştırılması
Bu çalışma, kızgınlık gösteren ineklerde, kızgınlığın farklı devrelerinde elde edilen vagina ve serviks akıntılarında kızgınlığın erkeğe habercisi olan feromonlarm varlığını ve kimyasal yapısını araştırmak amacıyla planlanmıştır. Serviko-vaginal mukus örnekleri östrusta bulunan 104 inekten östrasun başında, ortasında ve sonunda rectal masaj yoluyla toplandı. Bu örnekler analizlere başlayıncaya kadar -15 ile -20 °C arasında muhafaza edildi. Serviko-vaginal mukusun Kloroform/Metanol, Dializ, Kolon Kromatografısi (Slica gel'le doldurulmuş) ve Dietüeter ile ekstraksiyon işlemleri yapıldı. Boğa denemesi sonucu müspet reaksiyon alman ekstraktların kimyasal yapısını tayin etmek için Ultraviyole-Visible, Infrared, $^1H$ -Nükleer Magnetik Rezonans ve Gaz/Kütle spektroskopisi kullanılarak spektrumları alındı. Serviko-vaginal mukus örneklerinde tespit edilen ortalama vizkozite 2.37+0.09, pH 7.46±0.06, kuru madde % 1.62+0.06, su % 98.37±0.07, Ca 10.06+0.06 mg/100ml, Na 12.11±0.05 mEq/1, K 118.04+0.66 mEq/l ve Mg 3.96 ±0.03 mg/100ml olarak bulunmuştur. Örneklerden yalnızca dietüeter ekstraksiyon işlemleri sonucunda elde edilen ekstraktlar boğa denemesi sonucu müspet netice vermiştir. Bu ekstraktların $^1H$ -NMR ve Gaz/kütle spektroskopileri sonucu dialkil substitüe benzen, tri substitüe fenol, ester, alken ve steroid fonksiyonel gruplarından 16 adet organik madde bulunmuştur. Sığır cervico-vaginal mukusunda feromonlarm bulunduğu, bunların dietüeter gibi düşük ısıda buharlaşan çözücülerle ekstrakte edilebileceği ve feromonlarm tayininde boğa denemesinin en güvenilir yol olduğu anlaşılmıştır.
Investigation of physicochemical properties and pheromones in cervical and vaginal secretions at different terms of estrous cows
The aim of this study was to determine the presence and biochemical structure of pheromones that are believed to function as a signal of estrus for male animals. Cervico-vaginal mucus samples were collected from 104 estrous cows by rectal massage at the early heat, mid heat and late heat of estrus. These samples were stored at between -15 to -20 °C until analysed. Chloform / methanol, diethyl ether extractions, dialysis, coloumn chromatography (filled with slica gel) of cervico-vaginal mucus were also performed.These samples were biological assed on bulls. Afterwards, to determine the chemical structure of the extracts from the positive samples, spectrums were determined using Ultraviole-visible, Infrared, $^1H$ -Nuclear Magnetic Rezonans and Gas-Mass spectrometers. In cervico-vaginal mucus samples viscosity, pH, dry matter, water content, Ca, Na, K and Mg concentrations were mean 2.37±0.09, 7.46± 0.06, 1.62+ 0.06 %, 98.37±0.07 %, 10.06±0.06, 12.11± 0.05mEq/l, 118.04 ± 0.66 mEq/1, 3.96 ± 0.03 mg/100ml respectively. After extractions by diethyleter, the samples still induced bull reaction and following spectroscopy, in these extracts from the functional groups of dialkil subsumed benzene, tri substitued phenol, ester, alkene and steroid, 16 organic substance were found.
