ANTİJEN UYARIMI SONRASI SIÇAN SERVİKAL LENF DÜĞÜMLERİNDE GERMİNAL MERKEZLER VE APOPTOTİK HÜCRELERİN İNCELENMESİ

Bu deneysel çalışmada, öncelikle lenf düğümlerinin bir antijenik uyarıma verdiği cevabın, korteksdeki germi-nal merkezlere özel bir önem verilerek incelenmesi amaçlanmıştır. Çalışmada 20 adet Wistar tipi erişkin rat kullanıldı. Bunların 12 tanesi deney grubu olarak ayrıldı ve servikal bölgeden deri altına 0,1 ml Salmonella paratyphi B-H antijeni verildi. Geri kalanı kontrol grubu olarak kullanıldı ve benzer şekilde 0.1 ml serum fizyolojik uygulandı. Uyarımı izleyen 1,2,3,4,5 ve 8. günlerde her iki grubun yüzeyel servikal lenf düğümleri çıkarıldı. Lenf düğümlerine rutin ışık ve elektron mikroskobu için tespit ve takip prosedürü tatbik edilerek, elde edilen parafin ve yarı ince kesitlere çeşitli boyama teknikleri uygulandı. Antijen uyarımı sonrası 3. gün-de primer foliküllerde lenfoblast artışı ile birlikte germinal merkez reaksiyonlarının başladığı gözlendi. Anti-jen uyarımı sonrası 5. günde germinal merkezler daha genişti ve çok sayıda mitoz aşamasında ve apoptotik hücreler gözlendi. Ayrıca ışık mikroskobu düzeyinde demirli hematoksilen boyamasında ilk kez zıt boya ola-rak pikrofuksin kullanılmış ve bu yöntemin germinal merkezlerdeki apoptotik hücrelerin belirlenmesinde oldukca başarılı olduğu ortaya çıkarılmıştır. Uyarımın 8. günü sekonder folikül ve germinal merkezlerindeki boyanabilen cisim makrofajı sayısı en yüksek düzeydeydi.
