Deneysel Diyabetik Ratların Beyin Dokusunda İrisin İmmünreaktivitesi Üzerine Losartanın Etkileri

Amaç: Hiperglisemik metabolik bir hastalık olan Diabetes Mellitus (DM)'un, enerji metabolizmasında değişiklik yaptığı bilinmektedir. DM'un santral sinir sisteminde patolojik değişiklere neden olduğu insanlarda ve deney hayvanlarında gösterilmiştir. Bu çalışmada deneysel DM oluşturulan sıçanların beyin dokusunda irisin ekspresyonu üzerine losartan'ın (LS) etkilerinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Çalışmamızda 18 adet ergin Wistar albino cinsi erkek sıçanlar kullanıldı. Deney hayvanları her grupta 6 hayvan olmak üzere 3 gruba ayrıldı. Kontrol grubuna 6 hafta boyunca herhangi bir işlem yapılmadı. DM grubuna 50 mg/kg tek doz streptozotosin (STZ) intraperitoneal (i.p) olarak uygulandı. DM+LS grubuna ise 50 mg/kg tek doz STZ i.p olarak verilmesinden sonra diyabetin oluşumunu takiben 10 mg/kg/gün LS oral olarak verildi. Deney sonunda sıçanlar servikal dekapitasyonla ötenazi edilip beyin dokuları çıkartılıp tespit edildi. Rutin histolojik takipler yapılarak dokular parafin bloklara gömüldü. Beyin dokuları avidin-biotin-peroksidaz yöntemi ile immünohistokimyasal irisin ekspresyonunun belirlenmesi için hazırlandı. İmmünohistokimyasal boyanmanın değerlendirilmesinde; immünreaktivitenin yaygınlığı ve şiddeti esas alınarak histoskor oluşturuldu. Bulgular: Kontrol grubu ile karşılaştırıldığında İrisin immünreaktivitesi DM grubunda istatistiksel olarak anlamlı bir şekilde azalmıştı. DM grubu ile karşılaştırıldığında ise irisin immünreaktivitesi DM+LS grubunda anlamlı olarak artmıştı. Sonuç: Bu çalışma ile diyabetik beyin hasarı üzerine LS'nin nöronal alanlardaki irisin immünreaktivitesi aracılığıyla olumlu etkiler yapabileceği kanaatine varılmıştır.

The Effects of Losartan on Immunoreactivity of Irisinon Brain Tissue of Experimental Diabetic Rats

Objective: Diabetes Mellitus (DM) is a hyperglycemic metabolic disorder that alters the energy metabolism. It was also observed that DM causes pathological changes on the central nervous system in both humans and experimental animal models. In this study; we aimed to investigate the effects of losartan (LS) on immunoreactivity of irisin on brain tissue of experimental diabetic rats. Material and Methods: In this study; 18 adult male Wistar albino rats were used. Experimental rats were divided into 3 groups consisting 6 rats in each group. In the control group; the rats didn't receive any drugs during the 6 weeks of study period. In the DM group; 50 mg/kg single dose streptozotocin (STZ) was administered intraperitoneally (i.p). In the DM+LS group; 50 mg/kg single dose STZ was administered i.p and 10 mg/kg/day LS was started orally after the confirmation of DM. At the end of the experiment, all of rats were euthanized by cervical decapitation and brain tissues were removed and fixed. The brain tissues were processed to determine immunohistochemical localization of irisin by using avidinbiotin-peroxidase technique. Histoscore was created according to the diffusiveness and severity of immunoreactivity in assessment of immunohistochemical staining. Results: There was a statistically significant decrease in the DM group in terms of irisin immunoreactivity compared to the control group. Irisin immunoreactivity was significantly increased in DM+LS group compared to DM group. Conclusion: We can conclude that LS can improve diabetic brain damage with the help of irisin immunoreactivity located in neuronal areas.
