Timokuinonun sığır periferal kan monükleer hücre proliferasyonuna etkisi

Amaç: Timokuinon (Tq), Nigella sativa'nın en önemli bileşeni olup memelilerin çeşitli hücreleri üzerinde olumlu biyolojik etkilere sahiptir. Bu çalışmada, sığırların periferal kan dolaşımı mononükleer hücreleri üzerine Tq'nin in vitro şartlarda etkisi araştırıldı.Gereç ve Yöntem: Farklı Tq yoğunluklarının sığır periferal kan dolaşımı mononükleer hücrelerinin yaşamına ve proliferasyonuna etkileri hücre kültürü (BrdU) ve ELISA metotları ile ve iki mitojenin (Concanavalin A; Con A ve Pokeweed Mitogen; PWM) varlığında ve yokluğunda araştırıldı. Bununla birlikte fenil boronik asit (FBA)'in muhtemel etkisi benzer şekilde ölçüldü.Bulgular: Tq, sığır kan dolaşımında bulunan mononükleer hücreler, kültür sıvısına 50 µg/mL miktarında eklendiğinde blastojenik etki gösterdi. 5 günlük 10?/mL yoğunluğunda hücre bulunan kültürlerde 50 µg/mL Tq, 0.485±0.06 düzeyinde blastogenetik cevaba sebep oldu (P

Effect of thymoquinone on proliferation of bovine peripheral blood mononuclear cells

Aim: Thymoquinone (Tq) is the most important component of Nigella sativa and has positive biological effects on various cells of the mammals. At present study, a possible effect of Tq on the large ruminant immune system was investigated by in vitro assays. Materials and Methods: Thus, the effects of different Tq concentrations on both survival and proliferation of bovine peripheral blood mononuclear cells were investigated by cell culture and ELISA methods in the presence or absence of two mitogens (Con A and PWM). In addition, phenyl boronic acid (PBA) was evaluated similarly.Results: Tq showed a blastogenic effect on peripheral mononuclear cells of cows when Tq was added to culture medium at a final concentration of 50 μg/mL.
