İNFEKSİYÖZ KERATOKUNJUNKTİVİTİS SEMPTOMLU SIĞIRLARDAN MOREXELLA BOVİS'İN İZALOSYONU VE KARAKTERİZASYONU
Bu araştırmada, infeksiyöz keratokonjunktivitis semptomlu 92 sığırın 9'unda Moraxella bovis saptandı. Sığırların hastalıklı gözlerinden alınan 107 örnekten, 11 M. bovis suşu izole edildi. İzole edilen suşların tümü besi yerinde düzgün kenarlı yuvarlak, yarı şeffaf, grimsİ beyaz ve hafifçe besi yerine gömülmüş koloniler oluşturdu . Subkültürde R tipi kolonilere hızlı bir değişim gözlendi. İncelenen tüm M. bovis suşları Gram negatif diplobasil olarak görüldü. Sıvı besiyerinde tüm suşlar hafif bulanıklık ve önemli derecede tortu oluşturarak üredi. Tüm suşlar penisilin mevcudiyetinde hücresel uzama, oksidaz, katalaz, 20 °C' de üreme, tween 80, kazein, koagüle serum ve jelatİn hidrolizasyon testleri yönünden pozitif, pigmentasyon, hareket, Mac Conkey agarda üreme, % 5 NaCl' e tolerans, oksidasyon-fermentasyon (0-F), karbonhidrat fern1entasyon, indol, hidrojen sülfid ve üre hidrolizasyon testleri yönünden negatifbul undu. İncelenen suşların 8'i koyun kanlı agarda hemolitik bulunurken, 6 'sı nitrat redliksiyon testinde pozitif sonuç verdi
The isolation and characterization of Moraxella bovis from cattle with the symptoms of infectious keratoconjunctivis
Moraxella bovis was detected in 9 out of 92 cattle with symptoms of infectious keratoconjuntivitis. Eleven strains of M. Bovis were isolated from 107 sanıp i es obtained from affected eyes of cattele. Colonies of all strains isolated were circular with a entire-edge, translucent, grayish-white and slightly indented into the medium. In subculture, rapid dissociation to rough colonies was observed. All M. bovis strains examined were appeared to be Gram-negative diplobacilli . In liquid medium, all strains produced slight turbidity and considerable deposit All strains produced positive results in the following tes ts: cellular elongation in the presence penicilline, oxidase, catalase, growth at 20 °C, tween 80 and cascin hydrolysis, coagulated senım and gelatine liquefaction. All stratins produced negative results in the following tests: pigmentation, motility, growth on Mac Conkey agar, tolerance to 5% NaCl, oxidation-fermentation (0-F), carbonhydrates fermentation, indole and hydrogen sulphide production and urea hydrolysis. OfM.bovis strains examined, eight were haemolytic on sheep blood agar and six were positive in nitrate reduction test.
