ANDROCTONUS CRASSICAUDA TOKSİN SCX8 İLE ASETİLKOLİN NİKOTİNİK RESEPTÖR ETKİLEŞMESİNDE GXXXG MOTİFİNİN ROLÜ
Amaç: Dünyanın en zehirli akrepleri arasında bulunan Androctonus crassicauda, Türkiye’de önemlienvenomasyon nedenleri arasında yer alır; ancak üzerinde yeterince farmakolojik çalışma bulun-mamaktadır. Bu türün venomunda bulunan SCX8 isimli toksinin sodyum kanalı özelliğindeki niko-tinik reseptörler ile etkileşmesinde GXXXG (GG4) motifinin rolü araştırılmıştır.Gereç ve Yöntem: Biyoinformatik çalışmalarda kullanılan uniprot.org veri bankasından protein bil-gileri indirilmiş, Linux işletim sistemi GNU/Bash komutları kullanılarak insan nikotinik reseptör veakrep SCX8 toksin bilgileri ekstrakte edilmiştir. Clustal omega programı ile nikotinik reseptörler veSCX8 toksin dizileri sıralanmış ve GG4 motif ilişkileri saptanmıştır. R programlama dili kullanılarakSCX8 toksinine ait amino asit dağılımları işlenmiş ve gösterilmiştir.Bulgular: SCX8 toksini ile insan nikotinik reseptörleri arasından sadece ACM3 alt biriminde GG4motif benzerliği bulunduğu görülmüştür.Sonuç: Türkiye’de önemli bir zehirlenme nedeni olan Androctonus crassicauda türünde bulunanSCX8 toksininin etki yöresi arasında ACM3 nikotinik reseptör alt biriminin rolü olabileceği, And-roctonus crassicauda ile envenomasyon sonucunda gözlenen enflamatuvar reaksiyonların, SCX8toksini ile ACHA3 nikotinik reseptör alt birimi arasındaki bu etkileşmeye bağlı olabileceği ve buspefisik etkisine bağlı olarak SCX8’in yeni ilaç geliştirilmesinde önemli rol oynayabileceği sonucunavarılmıştır.
The role of GXXXG motif on the interaction of SCX8 toxin of Androctonus crassicauda and acetylcholine nicotinic receptor.
Objectives: Androctonus crassicauda is one of the deadly scorpions of the world and one of the important cause of envenomation lacking sufficient pharmacological investigations. Objective of this study is to investigate the role of GXXXG (GG4) motif on the interaction of one of the toxins of this scorpion named SCX8 and nicotinic receptors. Materials and Methods: Data on toxins were downloaded from the uniprot.org database, extracted and processed using GNU/Bash commands of Linux operating system. Nicotinic receptor subunits and SCX8 toxin sequences were aligned using Clustal omega software. The amino acid distribution of the SCX8 toxin was depicted using the R programming language. Results: Presence of GG4 motif similarity was only found on ACM3 subunit of sodium channels and the toxin SCX8. Conclusion: ACM3 subunit of nicotinic acetylcholine receptor may be a site of action of the toxin SCX8 of Androctonus crassicauda, which is an important envenomation factor, and this specific interaction indicating the role of SCX8 on the inflammatory reactions due to envenomations of this scorpion venom and suggesting this toxin may have a role in the new drug development studies.
