Bazı fasulye genotiplerinin Bean common mosaic virus (BCMV) ve Bean common mosaic necrosis virus (BCMNV)’a dayanıklılık durumlarının kalitatif, kantitatif ve moleküler yöntemlerle belirlenmesi

2007 yılında ikisi tescilli olmak üzere beş fasulye çeşidi ve bir ıslah hattının BCMV ve BCMNV'ye karşı dayanıklılık düzeylerinin ve sahip oldukları dayanıklılık genlerinin kalitatif, kantitatif ve genetik olarak belirlenmesine çalışılmıştır. Bu amaçla her çeşitten beş bitki, BCMNV NL-3 ırkı ve BCMV NL-4 ırkı ile inokule edilerek çeşitlerin dayanıklılık düzeyleri simptomatolojik olarak incelenmiştir. Buna ilave olarak, DAS-ELISA yöntemi ile çeşitlerdeki virüs enfeksiyon düzeyi kantitatif olarak da ortaya konmuştur. Ayrıca, dominant I genine spesifik SW-13 ve resesif bc-12 genine spesifik SBD-5 SCAR markörleri ile PCR yöntemi uygulanarak çeşitlerin söz konusu genleri bulundurma durumu moleküler olarak belirlenmiştir. Zülbiye ve Özeren Şeker çeşitlerinin ve 4F-3260 ıslah hattının BCMV'ye karşı dayanıklılıkta rol oynayan dominant I genine, Akdağ çeşidinin NL-4 hariç BCMV ve BCMNV'nin bilinen bütün ırklarına karşı dayanıklılığı sağlayan resesif bc-22 genine, Özeren Şeker ve Özeren Barbunya çeşitlerinin ise BCMV'nin dört (NL-1, NL-7, US-2, NL-2) ve BCMNV'nin NL-8 ırkına karşı dayanıklılığı sağlayan bc-12 genine sahip olduğu tespit edilmiştir.

Determination of resistance to Bean common mosaıc virus (BCMV) and Bean common mosaic necrosis virus (BCMNV) in some common bean genotypes using qualitative, quantitative and molecular methods

In 2007, a total of five bean cultivars, two of which are commercial cultivars, and one breeding line were evaluated for presence of known resistance genes and resistance levels to BCMV and BCMNV using qualitative, quantitative and molecular methods. Five plants of each cultivars and breeding line were inoculated with NL-3 strain of BCMNV and NL-4 strain of BCMV, and all plants were evaluated for virus symptoms and vigor. Additionally, plants were tested by DAS-ELISA to evaluate the virus titer on non-inoculated trifoliate leaves quantitatively. Also, two SCAR markers, SW-13 linked to the dominant I gene and SBD-5 for the recessive gene bc-12 were used to screen these genes in those cultivars by PCR. Results showed that cultivars Zulbiye and Ozeren Seker and 4F-3260 breeding line posses the dominant I gene which is responsible for resistance to BCMV, and Akdag has the recessive gene bc-22 which confers resistance to all known strains of BCMNV and BCMV except NL-4. Cultivars Ozeren Seker and Ozeren Barbunya occurred to possess the recessive gene bc-12 which is responsible for resistance to four strains of BCMV (NL-1, NL-7, US-2, NL-2) and NL-8 strain of BCMNV.
