Tavukböbreğinde mast hücrelerinin dağılımı ve heterojenitesi

Bu çalışma tavukların (Gallus domes- ticus) böbreklerindeki mast hücrelerinin ışık mikroskobik düzeyde morfolojisi ve farklı tespit sıvıları kullanılarak mast hücre hetero- jenitesini araştırmak amacıyla yapıldı. Araş- tırmada, aynı koşullarda yetiştirilen 21 ve 42 günlük, her yaş grubunda 6 adet olmak üzere toplam 12 adet brolier tavuğun böbreğinden alınan örnekler materyal olarak kullanıldı. Doku örnekleri % 10’luk formol ve izotonik formaldehit asetik asit (IFAA) solüsyonlarında tespit edildi. Mast hücreleri için toluidin blue ve alcian blue/safranin O (AB/SO) kombine boya metodları uygulandı. Işık mikroskobik düzeyde yapılan incelemeler sonucunda mast hücrelerinin, yerleşim yerlerine göre yuvarlak, oval ve mekik şeklinde ve farklı büyüklüklerde olduğu görüldü. IFAA tespit sıvısı kullanıldı- ğında böbrekte, %10’luk formolde tespit edi- lenlere göre daha fazla sayıda mast hücresinin bulunduğu gözlendi. Işık mikroskobik düzey- de yapılan incelemeler sonucunda hem IFAA hemde formol ile tespit edilen dokularda mast hücreleri belirgin şekilde metakromazi göster- meleri ile ayırt edildi. AB/SO kombine boya metodu uygulanan kesitlerde AB(+), SO(+) ve AB/SO(+) (mikst) mast hücrelerine rast- landı. Formol tespiti yapılmış olan kesitlere farklı bir boya yöntemi olan alcian blue tetra- kis (methylpyridium chloride) boyası yapıldı. Alcian blue tetrakis’in mast hücre granüllerini AB/SO boyamasına oranla daha koyu olarak boyamış olduğu görüldü.

Age related distrubition and heteroge-nity of mast cells in the kidney of Gallus domesticus

Abstract: This study aimed to determine the morphology of mast cells of the kidney of Gallus domesticus at light microscopic level and determine the heterogeneity of mast cells based on different fixation solutions. In this study, grown under the same conditions total of 12 Gallus domesticus were divided into two group (21, 42) according to their ages and kidneys were removed follwing decapitation. Tissue samples were fixed in both 10% for- maldehyde and isotonic formaldehyde acetic acid (IFAA) solutions. Toluidine blue and al- cian blue /safranin O (AB/SO) combined sta- ining methods were applied for the mast cel- ls. As a result of the examination on the light microscopic level, it was observed that mast cells have spherical, ovoid and elongated, de- pending on their location, shapes and different sizes. It was founded that IFAA fixed tissu- es were contained more mast cells than those tissues fixed in the 10% formaldehyde. As a result of light microscopical examinations, tis- sues which were fixed with both formaldehy- de and IFAA fixation solutions mast cells were distinguished by significantly demonstrate metachromasia. With Alcian blue and safranin O staining, in the section from gallus domes- ticus kidney regardless of their location mast cells exhibited Alcian blue positive, safranin O positive and mixed reaction. Formaldehyde fixated sections were stained with a different staining method called alcian blue tetrakis (methylpyridium chloride). It was seen that al- cian blue tetrakis dye stained granules of mast cell denser than the AB/SO dye.
