Increasing the injured area on hybocotyl explants of flax (Linum usitatissimum L.) leads to high frequency callus-based shoot regeneration

Bu araştırmada, keten bitkisinin hipokotil eksplantlarında, epidermis tabakası soyularak yaralanan yüzeyin genişletilmesi ile Agrobacterium inokulasyonundan sonra transgenik olmaya aday sürgün frekansının artırılması amaçlanmıştır. Soyulmamış hipokotil eksplantlarında kallus ağırlığı, sürgün oluşturan hipokotil yüzdesi, eksplant başına sürgün sayısı ve petrideki sürgün sayısı sırasıyla 0.8 g, %40, 4.24 adet ve 16.96 adet olarak belirlenmiştir. Diğer taraftan, epidermis tabakası soyulduğunda, kallus ağırlığı, sürgün olußturan hipokotil yüzdesi, eksplant başına sürgün sayısı ve petrideki sürgün sayısı sırasıyla 1.35 g, %58.75, 8.15 adet ve 47.88 adete yükselmiştir.

Keten (Linum usitatissimum L.) hipokotil eksplantlarında yaralanan yüzeyin artırılması kallustan gelişen sürgün frekansını yükseltmektedir.

In this study, it was aimed to increase the frequency of shoots, which would be nominated to be transgenic after Agrobacterium inoculation, through expanding the wounding area by peeling the epidermis layer of hypocotyl explants of flax plants. Callus weight, percentage of hypocotyls producing shoots, shoot number per explant and total shoot number per petri dish were recorded as 0.8 g, 40%, 4.24 and 16.96, respectively in non-peeled explants. On the other hand, when the epidermis layer was peeled, callus weight, percentage of hypocotyls producing shoots, shoot number per explant and total shoot number per petri dish increased to 1.35 g, 58.75%, 8.15 and 47.88, respectively.
