Effect of $alpha$ -Keto acids against $H_2O_2$- and $NaN_3$ -Induced mutagenesis in different strainst of Salmonella typhimurium

Salmonella typhimurium TA98, TAİ00 ve TAİ02 suşlarında a-keto asitlerin, H2O2 ve NaN3'nin mutajenitelerine karşı koruyucu etkileri araştırıldı. H202 (340 ug/plak), hem TA98 hem de TA102 susunda mutajenik iken, NaN3 (1.5 ug/plak) sadece TA100 susunda mutajeniktir. Pürüvat (220-440 $mu$ g/plak) ve a-ketoglutarat'ın (335-600 ug/plak) H202'ye karşı koruyucu etkileri, %37- %65 olarak bulundu, $alpha$ -keto asitlerin, TA100 susunda $NaN_3$ tarafından indüklenen mutajenesise karşı koruyucu etkilerinin olmadığı saptandı. TAİ 02 susunda, NaN3 (1.5 $mu$ g/plak) ve $alpha$ -keto asitlerin (330-500 $mu$ g/plak) varlığında, $H_20_2$'nin (340 $mu$ g/plak) mutajenik etki göstermediği bulundu. Bu çapraz-adaptif cevap, TAİ 02 susunda DNA onarım sistemin sağlam olmasından kaynaklanmış olabilir. TA98 ve TAİ 00 suşlarında ise, $H_2O_2, NaN_3$ ve a-keto asitlerin yüksek konsantrasyonlarında kombinasyonlarının sitotoksik etki gösterdikleri bulundu. Çünkü bu suşlar, DNA onarım sistemlerinde hasara sebep olan uvr B mutasyonu taşımaktadırlar.

Salmonella typhimurium' un farklı suşlarında $H_2O_2$ ve $NaN_3$ ile indüklenmiş mutasyonlara karşı $alpha$ -keto asitlerin etkisi

The protective effects of $alpha$ -keto acids on the mutagenicity of $H_20_2$ and $NaN_3$ was investigated in Salmonella typhimurium TA 98, TA 100, TA 102. H202 (340 $mu$ g/plate) was mutagenic to the strains TA 98 and TA 102 while NaN3 (1.5 $mu$ g/plate) was mutagenic only to TA 100.The extents of protective effect of pyruvate (220-440 $mu$ g/plate) and $alpha$ -keto glutarate (335-600 ug/plate) against $H_20_2$ were observed to be 37%- 65% in TA 98 and TA 102. a-keto acids were ineffective against $NaN_34 induced mutagenicity in TA 100. H202 (340 $mu$ g/plate), in the presence of $NaN_34 (1.5 $mu$ g/plate) and $alpha$ -keto acids (330-500 $mu$ g/plate), was found to be nonmutagenic to the strain TA102. This cross-adaptive response might have been due to the intactness of the DNA repair system in TA 102. The above combination of $H_20_2, NaN_3$ and $alpha$ -keto acids were found to be cytotoxic in TA 98 and TA 100, possibly because these strains carry uvr B mutation causing defect in their DNA repair system.
