Ratlarda bleomisin ile oluşturulan akciğer fibrozisinde erdosteinin inflamasyon ve fibrozis üzerine etkileri

Bleomisin (BLM) uygulaması ile ratlarda akciğer fibrozisi oluşturarak, akut inflamatuvar ve fibrotik fazlarda akciğer dokusunda ve bronkoalveolar lavaj (BAL) sıvısında bazı kemokin ve sitokinlerin düzeyini, akciğer dokusunda oluşan histopatolojik değişiklikleri ve antioksidan etkisi olan erdosteinin akut inflamatuvar değişiklikler ve fibrozis üzerine etkilerini araştırmayı amaçladık. Çalışmaya Wistar tipi 45 erkek rat alındı. 0.günde; kontrol grubuna (grup 1, n=15) intratrakeal (i.t.) salin, BLM (grup 2, n=15) ve erdostein grubuna (grup 3, n=15) i.t. BLM (7.5 U/kg) uygulandı. Erdostein grubuna BLM uygulamasında 2 gün önce peroral 10 mg/kg/gün erdostein başlandı. Tüm gruplarda 0., 14. ve 29. günde 5’er adet rat sakrifiye edilerek BAL yapıldı. BAL’da malondialdehit (MDA), makrofaj inflamatuar protein 1 alfa (MIP-1α), MIP-2 düzeylerinin ölçümü ve inflamatuvar hücre sayımı, akciğer dokusunda hidroksiprolin ölçümü ve histopatolojik inceleme yapıldı. Effects of erdosteine on inflammation and fibrosis in rats with pulmonary fibrosis induced by bleomycin

We aimed to investigate the levels of some chemokines, inflammatory cell counts in bronchoalveolar lavage fluid (BALF), histopathological changes in lung tissue, to determine the effect of erdosteine on acute inflammatory changes and fibrosis in a rat fibrosis model induced by Bleomycine (BLM). Forty-five Wistar male rats were taken into the study. On day 0, intra tracheal (i.t.) saline to control group (group 1, n=15), i.t bleomycine 7.5 U/kg to bleomycine (group 2, n=15) and erdostein group (group 3, n=15) was administered. In group 3, oral erdostein (10 mg/kg/day) was applied two days before bleomycine. On day 0, 14, and 29. five rats in each groups were sacrificed, BALF was performed. Malonyldialdehyde (MDA), macrophage inflammatory protein 1 alpha (MIP-1α), MIP-2 levels in BALF, hydroxyproline levels in lung tissue were measured. Histopathological examination was performed.
