Thiacloprid ve D-Tubokurarin'in Rana ridibunda Gastrokinemius Kası Üzerine Toksik Etkileri III: Oksidatif Potansiyel

Bu çalışmada neonikotinoid bir insektisit olan thiacloprid ve antagonisti d-tubokurarin'in kurbağa gastrokinemius kasında, tiyobarbitürik asit reaktif madde düzeyleri ve katalaz enzim aktivitesi üzerine etkileri araştırılmıştır. Deneylerde 35 adet kurbağa kas preparatı kullanılmıştır. Gastrokinemius kası izole edildikten sonra, 120 dakika boyunca 250, 25, 2,5 ve 0,25mg/L olmak üzere thiacloprid'in 4 farklı konsantrasyonuna maruz bırakılmıştır. Ayrıca, izole kaslar 2,5 mg/L thiacloprid ile 80 mg/L d-tubokurarin karışımı ve 0,25 mg/L thiacloprid ile 8 mg/L d-tubokurarin karışımı ile muamele edilmiştir. Kontrol grubundaki kas dokuları ise, 120 dakika boyunca Ringer çözeltisi içerisinde bekletilmiştir. Toxic Effects of Thiacloprid and D-Tubocurarine on Rana ridibunda Gastrocnemius Muscle III

In this study, the effects of neonicotinoid insecticide thiacloprid and its antagonist d-tubocurarine on the amount of thiobarbituric acid reactive substances and their effects on catalase enzyme activity was investigated in frog gastrocnemius muscle.In the experiments 35 frog muscle preparations were used. The isolated gastrocnemius muscle was subjected to four different concentrations of thiacloprid (250, 25, 2.5 ve 0.25mg L-1) for 120 minutes. The muscles were also treated with a 2.5 mg L-1 thiacloprid and 80 mg/L dtubocurarine mixture, with 0.25 mg L-1 thiacloprid and 8 mg L-1 d-tubocurarine mixture. The muscle tissues in the control group were maintained in Ringer's solution for 120 minutes. Each concentration group was studied with an equal number of preparations (n=5). Based on the research results, it was determined that 250, 25, 2.5 ve 0.25mg L-1 thiacloprid and the mixture of 0.25 mg L-1 thiacloprid and 8 mg L-1 d-tubocurarine decreased the amount of thiobarbituric acid reactive substances of the striated muscle tissue compared to the control group (P<0.05). 250 (P<0.001) and 25 mg L-1 thiacloprid (P<0.05) decreased the catalase enzyme activity in muscle tissue. This study provides important data for the potential of thiacloprid and d-tubocurarine creating oxidative stress in skeletal muscle, for assessing the possible effects on non-target organisms and for assessing their environmental risks.
