RATLARDA KARBON TETRAKLORÜR İLE OLUŞTURULAN KRONİK KARACİĞER HASARI ÜZERİNE NAR ÇEKİR-DEĞİ YAĞININ ETKİSİ VE KASPAZ AKTİVİTESİ İLE HEPATİK APOPTOZİSİN BELİRLENMESİ

Bu çalışmada, karbon tetrakloru r (CCl4) ile kronik kara-ciğ er hasarı oluşturulan ratlarda nar çekirdeğ i yağ ının (NÇY) karaciğ er dokusunda oluşabilecek lezyonlar u ze-rine koruyucu etkisi ve biyokimyasal değ erleri araştırıl-mıştır.Çalışmada 32 adet, her ğrupta 8 rat olmak u zere 4 ğruba ayrıldı. İ lk ğrup kontrol olarak belirlendi. İ kinci ğruba 12 hafta her ğu n 2 ml/kğ dozunda NÇY ğavaj ile verildi. Ü çu ncu ğruba dilu e edilen 0.2 ml/kğ iki doz CCl4 her hafta 3 ğu n ara ile intraperitoneal olarak enjekte edildi. Do rdu ncu ğruba 2 ml/kğ dozunda NÇY 12 hafta her ğu n, ğavaj ile verilirken her hafta 3 ğu n ara ile 2 doz CCl4 uyğulandı.Kontrol ğruplarında normal karaciğ er doku yapısı ğo zlenirken; CCl4+NÇY ğrubu hepatositlerde CCl4 ğrubuna ğo re daha az yoğ unlukta makro ve mikro-veziku ler yağ vakuolleri ğo ru ldu . NÇY uyğulamalarında histopatolojik değ işiklikler kısmen CCl4 ğrubuna ğo re kısmen azalırken, kaspaz 3 ve 9 aktivitesi de sınırlı sayı-da azaldı. Kronik olarak CCl4 uyğulanan ğruplarda se-rum ALT aktivitesi ve karaciğ er MDA du zeyleri o nemli du zeyde artış ğo sterirken, NÇY uyğulamaları istatistiki o nemde etkilenmedi.Kronik CCl4 ile NÇY verilen deney ğruplarında karaciğ er hasarı u zerinde biyokimyasal veriler ve histopatolojik lezyonlar u zerine iyileştirici yo nden anlamlı bir etkisi ğo zlenmedi.

THE EFFECTS OF POMEGRANATE SEED OIL ON CHRONIC LIVER DAMAGE INDUCED CARBON TETRACHLO-RIDE IN RATS AND DETERMINATION OF HEPATIC APOPTOSIS AND CASPASE ACTIVITY

İn this study, the protective effect, and biochemical val-ues of pomeğranate seed oil (PSO) on liver tissue le-sions in rats with a chronic liver injury caused by car-bon tetrachloride(CCl4)were investiğated.Thirty-two rats were divided into 4 ğroups as 8 rats in each ğroup. The first ğroup was determined as control. The second ğroup received PSO ğavağe at 2 ml/kğ daily for 12 weeks. İn the third ğroup, 0.2 ml/kğ two doses of CCl4 were injected intraperitoneally at 3-day intervals in each week. The fourth ğroup received PSO at 2 ml/kğ daily by ğavağe for 12 weeks, while 2 doses CCl4 at 3 days intervals in each week. While normal liver tissue structure was observed in the control ğroups; Macro and microvesicular fat vacuoles were less common in CCl4+PSO ğroups hepatocytes than CCl4 ğroup. İn PSO applications, histopatholoğical chanğes were partially decreased compared to the CCl4 ğroup, while caspase 3 and 9 activity decreased to a limited number. Serum ALT activity and liver MDA levels were siğnificantly increased in the chronic CCl4 treated ğroups, while PSO applications were not siğnificantly affected. İn the ex-perimental ğroups treated with chronic CCl4 with PSO, no siğnificant effect was observed on biochemical data and histopatholoğical lesions on liver damağe.
