MELATONİN VE C VİTAMİNİNİN KRONİK ALKOLİK SIÇANLARIN SEREBELLUM DOKUSUNDAKİ HASAR VE eNOS İMMUNREAKTİVİTESİ ÜZERİNE ETKİLERİ

Bu çalışmada etanolün serebellumda oluşturduğuhasar ve eNOS ekspresyonu üzerine melatonin ve Cvitamininin amaçlandı.Çalışmada 24 adet 200-250 gr ağırlığında Wistar cinsiergin erkek sıçan kullanıldı. Sıçanlar rasgele olarakdört gruba ayrıldı. Birinci grup kontrol (n: 6); ikincigrup deney süresince (28 gün) alkol içeren sıvı diyetalan (n: 6); üçüncü grup deney boyunca alkol içeren sıvıdiyet ile birlikte 40mg/kg/gün intraperitoneal C vitaminialan (n: 6); dördüncü grup deney süresince alkol içerensıvı diyet ile birlikte 4mg/kg/gün intraperitonealmelatonin (n: 6) alan sıçanlardan oluşturuldu.Deney süresince sadece alkol alan gruptaki sıçanlarınserebellum dokularında gangliyoner tabakada purkinjehücrelerinde dejenerasyon ve hücre sayılarında azalmagözlendi. Melatonin ve C vitamini uygulanan gruplardapurkinje hücrelerindeki dejenerasyon oranının azaldığıayırt edildi. Kontrol grubu sıçanlarda, endotelyal nitrikoksit sentaz immunoreaktivitesi için yapılan boyamadamoleküler ve granüler tabakalarda (++) boyanma,purkinje hücrelerinde ise (+++) boyanma gözlendi.Alkol alan grupta dejenere olan purkinje hücrelerindeendotelyal nitrik oksit sentaz immunoreaktivitesigözlenmedi. Melatonin ve C vitamini uygulanangruplarda ise (+++) boyanma mevcuttu.Sonuç olarak kronik alkol tüketimi serebellumdokusunda hasara yol açmakta. Bu hasar melatonin veC vitamini ile kısmen düzelmektedir

Effects of Melatonin and Vitamin C on Damage and Expression of Endothelial NOS in Cerebellum of Chronic Alcoholic Rats

The aim of this study was to determine thealterations in cerebellum caused by alcoholconsumption and the effects of melatonin and vitaminC on these changes, in rats.Twenty-four adult male Wistar rats weighting 200250g were used in this study. Rats were divided intofour groups. The first group served as control (n:6).The second group was the ethanol treatment for 28days (n:6). The third group was given ethanol andsupplemented (intraperitoneally) (n:6). The fourth group was givenethanol and supplemented with 4 mg/kg/daymelatonin (intraperitoneally) (n:6).Light microscopic examinations revealed Purkinjecell degeneration and decrease in the number ofPurkinje cells in ganglionic layer of cerebellum. Inmelatonin and vitamin C treated groups, degenerationin Purkinje cells was less than the other groups. In thecontrol group, endothelial nitric oxide synthaseimmunoreactivity was (+++) in Purkinje cells, and(++) in granular and molecular layer. No endothelialnitric oxide synthase activity in degenerated Purkinjecells of alcohol group was observed. In melatonin andvitamin C treated groups, eNOS immunoreactivitywas (+++).Present results indicated that chronic alcoholconsumption increased lipid peroxidation andespecially melatonin and vitamin C administrationproduced in some degree protection against alcoholinduced damage in cerebellum
