Uyanık ratlarda koroner ligasyon sonucu oluşan aritmiler ve ölüm oranı üzerine glibenklamide (ATP-bağımlı potasyum kanal blokeri) ve metprolol'ün (B-1 kardiyoselektif adrenerjik bloker) birlikte etkisi
Glibenklamide, iskemik hücrelerde hücre içi ATP azalmasına bağlı olarak açılan potasyum kanallarını bloke eden bir sulfonilüre bileşiğidir. Koroner ligasyon sonrası aritmi şiddeti ve ölüm oranını azaltıcı etkisi ortaya konmuştur. Ancak bu ilacın aynı zamanda iskemiyi artırıcı etkisi de saptandığından, antiaritmik etkisi tartışmalıdır. The combined effect of glibenclamide (ATP-dependent potassium channel blocker) and metoprolol (cardioselective adrenergic B-1 blocker) on the arrhythmias and survival rate following coronary arter ligation in conscious rats
Glibenclamide is a sulfonilüre compound that blocks the potassium channel that opens in response to decrease intra-cellular ATP in ischemic cells. It has been found that it decreases the severity of arrhythmias and the survival rate after the coronary ligation. Since it is also known that it increases ischemia, the antiarrhythmic effect of glimenclamide is controversial. On the other hand, metoprolol has been known to decrease the ischemia. That is why, it can be thought that the anthyarrhythmic effects of glibenclamid may be further potentiated by metoprolol. The coronary ligation was produed in conscious animals and ECG was recorded during the 15 minutes following coronary ligation. The type, length and score of arrhythmias were determined by evaluating the ECG for each animal. The survival rate at the end of 15 th min of coronary ligation in animals treated with glibenclamide and metoprolol combination, were increased significantly (It was %9 in control; %62 in combination group). The score of arrhythmia was also decreased significantly (p<0.05) in this group. As a result, it is observed that metoprolol increased the cardioprotective effects of glibenclamide. This effects may be due to metoprolol that it may lead to decrease myocardial oxygen consuming of normal and ischemic myocardium. The cardioprotective effects of glibenclamide alone not to be observed similar to the previous studies, may be caused by the age and body weight of rats used in this study. That is why, it will be valuable to make research to reveal the effects of different age and body weight of animals on the arrhythmias following coronary ligation.
