Köpeklerde deneysel akut parasetamol (Asetaminofen) toksikasyonu ve sağaltımında N-Asetilsistein'in etkinliği

Bu çalışmada köpeklerde akut parasetamol (Asetaminofen) toksikasyonunda klinik ve hematolojik bulguların belirlenmesi ve oral N-Acetylcysteine'in (NAC) sağaltıcı etkinliğinin araştırılması amaçlandı. 500 mg/kg dozda parasetamol ile zehirlenen toplam 11 köpeğin 5'i toksikasyon ve 6'sı sağaltım grubu olarak ayrıldı. Sağaltım grubundaki köpeklere toksikasyondan sonraki 4. saatten itibaren oral NAC verilirken, toksikasyon grubundakilere herhangi bir uygulama yapılmadı. Tüm köpeklerden toksikasyon öncesi (0) ve sonrasındaki 4, 8, 24,48, 96 ve 288. saatlerde klinik (mukozaların kontrolü, vücut ısısı, nabız ve solunum sayıları vb.) ve hematoloji (RBC, MCV, HCT, WBC, Hb, MetHb, pH, pCO2, pO2) HCO3, Sat O2, BD, AST, ALT, ALP, total ve direkt bilirubin, total protein, kreatinin ve prothrombin zamanı) muayeneler yapıldı. Experimental acute paracetamol (Acetaminophen) toxication and the therapeutic effect of N-Acetylcysteine in dogs

In this study, it was aimed to determine of clinical and haematological findings of acute paracetamol (Acetaminophen) toxication and to investigate the therapeutic effect of oral N-Acetylcysteine (NAC) application in dogs. Dogs toxicsted with 500 mg/kg Acetaminophen were divided into the toxication (5 dogs) and therapy (6 dogs) groups. While NAC was administred orally to therapy group apart from 4th hour of toxication, no attempt was made to toxication group. Clinical (Mucosa controls, body temperature, pulsation and respiration rate) and hematological (RBC, MCV, HCT%, WBC, Hb, MetHb, pH, $pCO_2, pO_2, HCO_3$ , Sat $O_2$, BD, AST, ALT, ALP, total and direct bilirubin, total protein, creatinine and prothrombin time) investigations were made before (0) and after toxication on the hours 4, 8, 24, 48, 96 and 288. In toxication group, methemoglobine at 4, 8 and 24, AST and ALP at 8, ALT at 48 and 96th hours of toxication were found higher (p<0.05, 0.01, 0.001) than 0 hours, whereas WBC at 4, MCV at 8 and 48., pO2 and SatO2 at 8, pCO2 at 24 and 288, PTT at 96 and 288 and HCO3 at 288th hours of toxication were low (p<0.05, 0.01, 0.001) from 0 hours. Following NAC administration, in therapy group Methemoglobine, AST, ALT, BD and PTT values were low (p<0.05, 0.01), while pCO2 and HCO3 values were high (p<0.05, 0.01, 0.001) in respect to toxication group. As a result, it was concoluded that beside the other clinical and haematological changes, the most characteristic findings of acute acetaminophen toxication in dogs were methemoglobinemia and increasing serum liver enzyme levels and additionally oral NAC administration was useful therapeutic method to prevent liver and erytrocytes from the toxic effect of acetaminophen.
