Kıvırcık koçlarinda donma ve eritme sonrasi spermatolojik özellikler üzerine mevsimin etkisi

Bu çalışmanın amacı dondurulmuş koç spermasını eritme sonrası mevsim içi ve mevsim dışı spermatolojik , özellikler açısından karşılaştırmaktı. Çalışmada 6 Kıvırcık koç kullanıldı. Koçlardan elektroejakülatörle sperma ' alındı ve bireysel spermatolojik özellikleri saptandı. Çalışma aşamasında alınan koç sperması pooling yapıldı ve süt tozu-yumurta sarısı-fruktoz sulandırıcısı ile 1:1 oranında sulandırılarak pellet yöntemine göre -79 °C 'da donduruldu. Pelletler % 2.9'luk Sodyum Şitrat-Dihitrat solüsyonunda 37 °C 'de eritildi ve spermatolojik özellikler saptandı. Belirtilen işlemler hem mevsim içi, hem de mevsim dışında tekrarlandı. Donma öncesi pooling aşamasında hacim hariç, diğer spermatolojik özellikler donma aşamasına kadar (sulandırma, soğutma, gliserolizasyon, ekilibrasyon) mevsimden etkilenmedi- Hacim ise mevsim içinde daha yüksek olarak saptandı (P< 0.05). Mevsim içinde eritme sonrası motilite % 52.60±10.97 olarak bulundu. Morfolojik olarak % 49.60±14.58 akrozom ve toplam % 50.03±14.52 bozukluk saptandı. Mevsim dışında % 35.5±12.33 motilite bulundu. % 55.41±11.66 akrozom ve toplam % 55.24±12.59 morfolojik bozukluk tespit edildi. Mevsim içinde motilite, mevsim dışına göre daha yüksek olarak saptandı (P< 0.05). Akrozoma ait ve toplam morfolojik bozukluklar ise mevsim dışında daha yüksek olarak bulundu (P< 0.05). Sonuç olarak, koç spermasında mevsimin etkisi donma ve eritme sonrası daha belirgindi ve mevsim içinde alınan koç sperması, dondurulabilme başarısı yönünden daha üstün bulundu.

The effect of season on spermatological characteristics of frozen-thawed semen in Kivircik rams

The aim of this survey was to compare breeding and anoestrus season in view of the spermatoiogicai .char¬acteristics in frozen - thawed semen in rams. In this study, 6 Kıvırcık rams was used as material. The ram semen was collected by electroejaculator and spermatoiogicai characteristics of each ram were determined. In the stage of experimental studies, collected ram semen was pooled in a tube (pooling) and diluted with skimmed milk powder-egg yolk-fructose extender at the ratio of 1:1. After then, extended semen was frozen at -79 °C by the method of pellet. The pellets thawed in a 2.9% /sodium citrate solution at 37 °C and spermatoiogicai characteristics were examined. Above mentioned processes "were repeated in both breeding and anoesturs season. ' At the all stages of prior to freezing (dilution, cooling, gliserolisation, equilibration), season was uneffected on spermatoiogicai characteristics, except for volume of pooled semen. However, the volume of pooled semen in breeding season was higher than anoestrus season (P<0.05). In breeding season, post-thaw motility was 52.6%+0.97. The acrosomal abnormalities and total morphological abnormalities were 49.60%±14.58 and 50.03%±14.52 respectively. In anoestrus season, post-thaw motility was 35.5%±12.33. The acrosomal abnormalities and total morphological abnormalities were 55.41 %±11.66 and 55 24%±12.59, respectively. Post-thaw motility in breeding season was higher than anoestrus season (P<0.05). In anoestrus season, acrosomaf and total morphological abnormalities were higher than breeding season (P<0.05). It is concluded that the spermatoiogicai characteristics of frozen semen is improved when collected during active breeding season as compared to semen during the period of sexuel quiescence, and the effect of season was more remarkable on frozen-thawed spermatoiogicai characteristics in rams.
