Inhibitory effect of hepatectomized (35%) mice serum upon the growth of L-cells

Erişkin memeli karaciğer hücreleri çeşitli fonksiyonları yapmak üzere farklılaştığından bölünmezler. Fakat, karaciğerin bir kısmı kesilerek çıkarıldığında, yani bir parsiyal hepatektomi (PH) operasyonu uygulandığında, karaciğer hücreleri hızla çoğalmaya başlarlar. Parsiyal hepatektomiden sonra hücre çoğalmasının başlatılmasını stimule eden ve karaciğerin büyümesine neden olan bir faktör saptanmıştır. Humoral faktör olarak adlandırılan bu faktörün hepatektomili hayvanların kan serumunda bulunduğu da ortaya konmuştur. Çalışmada, normal ve %35 parsiyal hepatektomi operasyonu yapılan farelerden serum elde edilmiş ve bu serum L-hücrelerinin yaşama ortamlarına ilave edilmiştir. Farklı günlerde her gruptaki canlı hücre sayılan ve işaretlenme indeksleri saptanmıştır. Elde edilen sonuçlar, %35 parsiyal hepatektomi uygulanan hayvanlardan elde edilen serumun, L-hücrelerinin büyümesini dördüncü günden sonra inhibe ettiğini göstermiştir.

Liver cells of mature mammals can not be divided since they are differentiated in order to carry out various functions. But when a part of the liver is cut out (partial hepatectomy=PH) the hepatic cells begin to multiply rapidly so make the regeneration of the liver possible. A factor has been determined which stimulates grown of the liver after the partial hepatectomy. This factor is called humoral factor and found in the blood sera of hepatectomized animals. In this study, sera are obtained from normal and 35% hepatectomized mice and this sera was added to medium of L-cells. In different days, number of living cells and labelling index were investigated. In conclusion, this study shows that sera obtained from 35% partially hepatectomized mice has an inhibitory effect on the growth of the cells, after the fourth day.
