DNA İzolasyonunda fenol-kloroform yerine potasyum asetat kullanımının DNA miktarı ve kalitesi üzerine etkisi
Bu çalışmanın amacı polimeraz zincir reaksiyonu (PZR) işlemlerinde kullanılmak üzere kandan genomik DNA izolasyonu amacıyla fenol-kloroform yöntemi yerine potasyum asetat çözeltisi kullanımının elde edilen DNA miktarı ve kalitesi üzerine etkisini araştırmaktır. Bu amaçla Kilis keçilerinden toplanmış olan 13 adet kan örneği kullanılmıştır. Her bir örnekte proteinaz K uygulamasını ardından fenol-kloroform ile DNA izolasyonu veya potasyum asetat ile çöktürme işlemi uygulanmıştır. Elde edilen DNA çözeltilerinin konsantrasyonları ve saflığı sırasıyla 260 nm ve 260nm/280nm de spektrofortometre ile ölçülmüştür. Fenolkloroform ve potasyum asetat grubunda ortalama DNA konsantrasyonu sırasıyla 0,4185±0,2368 ve 0,2753±0,0846 bulunmuştur. Absorbans oranları ise fenol-kloroform ve potasyum asetat grubunda sırasıyla ortalama 1,6380±0,1817 ve 1,7435±0,0659 olarak tespit edilmiştir. Gruplar arasındaki fark istatistiksel açıdan önemli bulunmuştur. Sonuç olarak DNA izolasyonunda proteinleri ortamdan uzaklaştırmak amacıyla potasyum asetat kullanıldığında fenolkloroform grubuna göre daha düşük düzeyde DNA elde edilmiş olsa da yeterli miktar ve kalitede DNA elde edilebildiğinden araştırıcının sağlığı ve çevreye zararlı olan fenol yerine potasyum asetatın DNA izolasyonunda kullanılabileceği kanaatine varılmıştır.
Effect of using potassium acetate instated of phenol-chloroform for DNA isolation on DNA yield and quality
The objective of this study was to investigate the effect of using potassium acetate instead of phenol-chloroform on the yield and quality of DNA used for polymerase chain reaction (PCR) assays. For this purpose 13 blood samples collected from Kilis goats were used. After proteinase K digestion, phenol-chloroform extraction or treatment with potassium acetate was performed on aliquots of each sample. Concentration and purity of DNA samples were measured at 260 nm and 260nm/280nm by using a spectrophotometer, respectively. Concentration of DNA in phenol-chloroform and potassium acetate groups were found to be 0.4185±0.2368 and 0.2753±0.0846 respectively. Absorbance ratio values of phenol-chloroform and potassium acetate groups were 1.6380±0.1817 and 1.7435±0.0659 respectively. Differences between groups were statistically significant. The results suggested that sufficient amount of DNA were obtained by using potassium acetate in order to remove proteins. Therefore potassium acetate can be used instead of phenol-chloroform which is more hazardous for the health of researchers or for the environment, although DNA yield obtained by using potassium acetate was lower than that obtained by using phenol-chloroform.
