Yurdumuz Sığır Leptospirosis'ine Karşı Etkin Bir Aşı Hazırlanması Üzerine Çalışmalar
Leptospira grippotypthosa sera grubundan yerli Kurşunlu suşu
kullanarak, manovalan numune aşı hazırlandı. Aşıda % 0, 1 formol,
% 0,01 merthioilate ile % 10 .Al(OH)3 kullanılmıştır . Aşı tohum kültürü
Johnson vasatında 30°C'de 7-10 gün inkube edilerek aşı üretildi.
Kültürden Hawslkley 'Thoma lamı kullanarak germ sayımı yapıldı
ve 7.2 x 10-8 germ/mL bulundu. Aşının sterilite, leptospiral
viability, toksisite testleri sırası ile uygulandı. Test sonuçları güvenilir
limitler içinde bulundu.
Hamsterlerdeki potency testi ıçın 10 adetlik üç grup 3 aylık
hamster alındı. 21 gün ara ile iki gruba 1/100 ve 1/400 sığır dozu
miktarında aşı inokule edildi. Aşılı hamsterler ikinci aşılamadan
15 gün sonra 10 adet aşısız kontrol hamsterle birlikte her üç grup
hamster 3 x 10-8 germ /mıl. pathogen L. grippotyphosa kültürünün
1000 LD50/0.2 ml. ile eprüve edildiler. Eprüvasyodan sonra 1/100
sığır dozu ile aşılı 1 inci hamster grubu hayatta kaldı. 0/ 10, 1/400
sığır dozu tatlbik edilen II inci hamster gruıbun üç tanesi öldü.
3/10 böylece birinci grup eprüvasyona % 100 dayandı. II. grup
ise % 70'te kalmıştır. Sığırlardaki bağışıklık testi hamsterlere paralel
olarak yapıldı, bu testte 24 dana kullanıldı. Danalar 5 ml. aşı
dozu ile 21 gün ara ile iki defa aşılandı. Aşılamadan altıncı ve onikinci
aylar sonu aşısız kontrol danalada birlikite 3 x 10-8 germ/ml.
hamsterden pasaj edilmiş pathogen kurşunlu kültürünün
1000 LD50/5 ml.'si ile deri altı ve gözde damlatılarak eprüve edildi .
Eprüvasyondan sonra idrarlarından ve böbreklerinden bakteriyel
kültürel ve histopatolojik muayeneler sonucu test danalarını leptospira
enfeksiyonuna karşı aşının en ,az % 83 oranında koruduğu
görüldü.
Saha denemesi olarak Çankırı'nın Kurşunlu, Çaylıca köyünde
433 baş sığır, 360 baş koyun aşılandı, aşılı hayvanda istenmeyen
lokal ve genel post vaccinal reaksiyonlara rastlanmadı. Aşı tohum
kültürü 'hamsterlerde her hafta pasıj suretiyle canlılığı devam ettirilirken,
buna paralel olarak aynı suş liquide nitrogen containerlerde
özel payet ve silikonlu tüp içinde % 10 gliserinde patojenitesini
kayıp etmeden en az iki senedir muhafaza edilmektedir.
Preparing a vaccine with local strains against bovine leptospirosis, Which has adsorbed to aliminium hydroxide and it has strong immunologic properties
A monovalent vaccine was prepared by using Kurşunlu strain
which is indigenous, belonging to the L. .grippotyphosa serological
group. In this vaccine 0,1 % formaldehye, 0,01 1% merthiolate and
10% Al(OH)3 had been used. Vaccinal seed germs have been incubated
at 30 c for 7-10 days and so the vaccine have been produced
.
. The germs counting was made by using Hawskley Thoma slide,
and the number of the germs was found as 7,2 x l0-8 germ/ml.
Leptospiral viability, toxicity and sterility tests have been applied
respectively. The results of the tests were found in reliable limits.
For potency test in hamsters, the animals, at the age of · 3
months were divided into 3 groups and each group was containing
ten hamsters. At interval of 21 days, the vaccine have inoculated
into two .groups with the amount of 1/100 and 1/400 cattle doses.
Vaccinated hamsters, after 15 days frorrn second vaccination
and togetlher with unvaccinated control hamsters have been challenged
witih 3 x 10-8 germ/m1. pathogen L.grippotyphosa cultures
wihich were 1000 LD50 /0,2 ml.
After challenge, first hamster group were alİve. These hamsters
had been vaccinated with 1/100 cattle dose. In the second
hamsters group, three animals were died. These hamsters had heen
vaccinated with 1/400 cattle dose.
And so tıhe fırst group was resistent to the challenge at ·the
rate of 100 %. The ratio was found 70 % at the second group. The
immunity test in the cattle, was made similar to hamsters. In ·this
test 24 calves were used. At the vaccination 5 ml. vaccine has been
used at interval 21 days. 6 and 12 months later from vaccination,
unvaccinated control calves and vaccinated calves were challenged
with 3 x 10-8 germ/mL In this challenge the culture which is called
Kurşunlu strain passaged from hamsters has been used as
1000 LD50 / 5 ml. subcutaneously and by dropping to eyes.
After challenge, bacteriological and histopathological examinations
from their urines and kidneys have been made. The results
showed that, the vaccine vas protective for calves at least, at ratio
of 83 %.
· As a field trial 433 cattle and 360 sheep have vaccinated in the
Çaylıca village/Kurşunlu. We didn't encounter undesirable postvaccinal.
Local and general, reactions.
The vaccinal seed culture has been maintained by serial weekly
passages in hamsters. Being paralel to this, the same strain with
10 % glycerine has been kept in the container which contain Liquide nitrogen , in the plastic pipette and silicon tubes. This strain
has been maintained for two years without lossing its pathogenity.
