Sağlıklı Boğaların Spermalarından Aerobik Bakteri İzolasyon ve İdentifikasyonu
Bu çalışmada, değişik ırklarda sağlıklı boğaların spermalarındaki bakteri
tipleri saptanmaya çalışılmıştır. Bunun için, Holstein, Esmer, Jersey, Simmental,
Brangus ve Limousin ırklarında toplam 55 sağlıklı damızlık boğa kullanılmış ve
steril sun'i vajenler yardımıyla sperma örnekleri toplanmıştır. Toplanan 55
örnekten uygun besiyerlerine ekimler yapılımıştır. Aeorobik koşullarda 37' C'
de 4 gün inkube edilen besi yerleri hergün kontrol edilerek üreyen kolonilerin
idenfikasyonu gerçekleştirilm i ştir.
Sperma örneklerinin yapılan aeorobik bakteriyolajik muayenelerinde toplam
61 bakteri izole edilmiştir. izolatların sayı ve oranları ; Bacillus sp. 20(%32.8),
Micrococcus sp. 11 (% 18.0), S. epidermidis 10(% 16.4), Streptococcus sp. 7
(% 11 .5) ve Coryne bacterium sp. 5(%8.2) olarak belirlenmiştir.
Sonuç olarak, sağlıklı boğaların spermasında potansiyel patojen nitelikli
bakterilerin de bulunabileceği düşünülerek damızlık boğaların veya bunların
spermalarının mikrobiyolojik kontrollerinin yapılmadan kullanılmaması ve hijyenik
önlemlerin alınması gereği anlaşılmıştır.
The identification and isolation of aerobic bacteri in semen of healthy bulls.
In this study, the types of bacteria in semen of healthy bulls of various
races were investigated. A total of 55 heatlhy buls of Holstein, Swiss Brown,
Jersey, Simmental, Limousin and Brangus races have been used and semen
samples have been collected by the help of sterile artificial vaginas.55 s'emen samples have been inoculated in convenient media. Agar plates
were incubated at 37 •c for 4 days in aerobic conditions w ith daily examinations
and the colonies grew on them were identified.
61 bacteria w ere isolated in aerobic bacterial examinations of semen samples.
Numbers and rates of isolated bacteria were established as Bacillus sp.
20 (32.8 %), Micrococcus sp. 11 (18.0 %), S. epidermidis 10 (16.4 %), E.coli
8(13.1 ), Streptococcus sp. 7 (11.5 %) and Corynebacterium sp. 5 (8.2 %),
respective Iy.
As a result, it was concluded that, potentially pathogen bacteria can be
found in semen of healthy bulls, so it has been understood that bulls ortheir
semen can not be used without a microbiological canfirmation and hygienic
protection should be taken .
