Koyun ve keçilerde küçük ruminantların vebası (Peste des petits ruminants) ve sığır vebası enfeksiyonlarının serolojik ve virolojik olarak araştırılması

Bu çalışmada klinik ve otopsi bulgularına göre şüpheli olarak değerlendirilen ı 7 koyun ve 6 keçi sürüsü örneklenmiştir. Bunlardan 15 koyun ve 5 keçi sürüsüne ait 206 kan serum örneği RPV ve PPRV antikorları yönünden test edilmiş, 8 koyun ve 2 keçi sürüsüne ait 38 izolasyon materyalinde RPV ve PPRV izolasyonu çalışmaları yapılmıştır. Kan serum örnekleri RP ve PPR C-ELISA testi ile test edilmiş ve RPV antikoru saptanmamıştır. Koyun serumlarının %87,95'i keçi serumlarının %90.00'ı PPRV antikorları yönünden pozitif bulunmuş, bu oranlar aynı zamanda PPR için morbidite oranı olarak kabul edilmiştir. Virus izolasyon çalışmalarında PDB ve PKB hücreleri kullanılmış, izolasyon materyallerinin 3'ünde PKB hücrelerinde CPE oluşturan bir etken tespit edilmiştir. Türkiye'de ilk defa CPE pozitif izolatlar I-ELISA testi kullanılarak PPRV olarak identifiye edilmiştir. İzolatların Vero-R hücrelerindeki CPE şekilleri, hücre yuvarlaklaşması, İs ve İn inklüzyon cisimcikleri ve sinsityal hücreler olarak belirlenmiştir. Gerçekleştirilen serolajik ve virolojik çalışmalar sonucunda, koyun ve keçilerde görülen şüpheli vakaların PPR olduğu, örnekle- nen şüpheli sürülerde RP enfeksiyonlarının bulunmadığı tespit edilmiştir. .

Serological and Virological Investigation of Rinderpest and Peste des Petits Ruminants in Sheep and Goats. In this study 17 sheep and 6 goats suspected flocks were sampled. Sera sample were taken 15 sheep, 5 goats flocks, and tested for antibody, 38 specimens from 8 sheep and 2 goats flocks, were used for virus isolation. Serum samples were test ed using RP and PPR C-ELI SA and all sera sample seronegative for RP antibodies, and %87,95 of sheep and %90,00 of goats sera sample were found seropositive for PPR antibodies. Virus isolation study was carried out in PCK and PLK cells and 3 specimens out of 38 formed CPE in PLK cells. All isolates were identified PPRV using I-ELISA. CPE of isolates characterised cell rounding, multinucleated syncytia formation, intracytoplasmic and intranuclear inclusion bodies in Vero-R cells monolayers. As a result in this study, suspected RP and PPR cases in sheep and goats were PPR and RP infection was absent in sampled flocks.
