Vimentin, sitokeratin, a-SMA ve desmin'in Yeni Zellanda tavşanı testis ve epididimisindeki immunohistokimyasal ekspresyonu

Çalışma vimentin, sitokeratin, desmin ve α-SMA (alfa smooth actin) gibi hücre iskeletinin yapısına giren bazı proteinlerin tavşan testis ve epididimislerindeki immunohistokimyasal lokalizasyonlarını ortaya koymak amacıyla planlandı. Araştırmada 15 adet sağlıklı erişkin erkek Yeni Zellanda tavşanından alınan testis ve epididimisler kullanıldı. İmmunohistokimyasal boyamalar için Strept-ABC boyama metodu uygulandı. Vimentin immunoreaktivitesi, seminifer tubüllerdeki Sertoli hücrelerinin perinüklear sitoplazmalarında, intertubüler alanlarda Leydig hücrelerinde, tubülus rektus ve rete testis epitelleri ile kan damarı endotellerinde belirlendi. Sitokeratin immunoreaktivitesi testiste belirlenmezken, epididimiste duktuli eferentis ve duktus epididimis epitellerinde gözlendi. α-SMA immunreaktivitesi, seminifer tubüllerdeki peritubüler myoid hücrelerde, rete testis, duktuli eferentis ve duktus epididimis epiteli altında uzanan myofibroblastlar ile damar duvarı ve tunika albugineada bulunan düz kas hücrelerinde belirlendi. Desmin için yapılan boyamalarda, desmin immunreaktivitesinin α-SMA’nınkine benzerlik gösterdiği dikkati çekti. Sonuç olarak, Sertoli hücreleri ile tubülus rektus ve rete testis epitel hücrelerinin vimentin intermediyer filamanlarını içermesi, tavşan testislerinde bu yapıların mezenşimal kökenli olduğunun belirtisidir. Ayrıca, α-SMA ve desminin aynı hücrelerde bulunması, spermatozoanların ve testikülar sıvının rete testis ve epididimise doğru taşınması ve duktus epididimis boyunca spermin geçişinde bu iki proteinin birlikte görev yaptığını göstermektedir.

The immunohistochemical expression of vimentin, cytokeratin, a-SMA and desmin in New Zelland rabbit testis and epididiymiş

This study was planned to determine the immunohistological localization of certain proteins that constitute to the cytoskelaton such as vimentin, cytokeratin, α-SMA and desmin in rabbit testis and epididiymis. The testis and epididymis patterns taken from fifteen healthy, adult, male New Zelland rabbits formed the material of our study. For the determination of the localization of vimentin, cytokeratin, α-SMA and desmin, a strept-ABC immunohistochemical staining procedure was applied. The vimentin immunoreactivity was determined in perinuclear cytoplasm of Sertoli cells, epithelium of tubuli recti and rete testis, and endothelium of blood vessels. The cytokeratin immunoreactivity was defined in the epithelium of ductuli efferentes and ductus epididiymis, but not in testis. α-SMA immunostaining was found in peritubular cells of the seminiferous epithelium, the myofibroblasts underlied the epithelium of rete testis, ductuli efferentes and ductus epididiymis, and the smooth cells in wall of vessels and tunica albuginea. In the staining for desmin, the desmin immunoreactivity was similar to that of α-SMA. In conclusion, the vimentin is found in the Sertoli cells and epithelium of tubulus rectus or rete testis suggest that those structures are derived from mesoderm. Futhermore, the α-SMA and desmin are present identical cellular structures exhibit that those proteins together serve to move the spermatozoa and testicular fluid towards rete testis and epididiymis.
