Sütçü İneklerin Kan ve Süt Serumlarında Bovine Leukosis Virus Enfeksiyonunun ELISA ile Araştırılması

Bu çalışma kan ve süt serumlarında Bovine Leukosis Virus (BLV)'a karşı gelişen antikorların tespit edilmesi ve kan serumu yerine süt serumu örneklerinin kullanılıp kullanılamayacağının belirlenmesi amacıyla yapıldı. Mart-Eylül 2010 tarihleri arasında Isparta'nın ilçelerindeki (Keçiborlu, Gönen, Şarkikaraağaç, Aksu, Yalvaç, Sütçüler, Atağbey, Eğirdir) özel işletmelerde bulunan klinik olarak sağlıklı görünümde BLV'ye karşı aşılanmamış 428 baş Holştayn ırkı (yaşları 2 ile 9 arasında) sağmal inekten serolojik incelemeler amacıyla kan ve süt örnekleri alındı. Aynı hayvanlardan elde edilen kan ve süt örnekleri BLV'ye spesifik antikor varlığını tespit etmek amacıyla indirekt Enzyme Linked Immunosorbent Assay (ELISA) ile incelendi. Araştırma sonucu kan serumu örneklerinin 86 adedi (%20.09) ve süt serum örneklerinin 39 adedi (%9.11) BLV antikorları yönünden pozitif bulundu. İlçeler dikkate alındığında ELISA-kan serum testi ile en düşük seropozitiflik %4 olarak Şarkikaraağaç'da tespit edilirken, en yüksek seropozitiflik ise %59.02 ile Gönen'de tespit edildi. ELISA-süt testi bulguları değerlendirildiğinde en düşük seropozitiflik %1.92 ile Aksu'da, en yüksek seropozitiflik ise %34.55 ile Yalvaç'da tespit edildi. Sonuç olarak ELISA-kan ve ELISA-süt testleri arasında ortanın altında uyum tespit edildiği (?= 0.214) için BLV'nin serolojik teşhisinde kan serumu yerine süt serumu örneklerinin kullanılamayacağı sonucuna varıldı

Investigation of Bovine Leukosis Virus Infection in Blood and Milk Serum in Dairy Cattle by ELISA

The aim of this study was to determine of antibodies against Bovine Leukosis Virus (BLV) in blood and milk serum and whether using milk serum instead of blood serum in serological tests for BLV could be preferred. In this study, 428 unvaccinated 2 to 9 years old clinically healthy Holstein breed dairy cows were examined from some private businesses in the districts of Isparta (Keçiborlu, Gönen, Şarkikaraağaç, Aksu, Yalvaç, Sütçüler, Atağbey, Eğirdir) between March 2010 and September 2010. Both blood and milk serum samples were collected from the same animal for serological investigation and all samples were examined by Indirect Enzyme Linked Immunosorbent Assay (ELISA) for BLV antibodies. Results showed that a total of 86 (20.09%) blood and 39 (9.11%) milk samples were detected positive for BLV antibodies. According to the districts, the lowest prevalance was detected as 4% in Şarkikaraağaç and the highest prevalance was detected as 59.02% in Gönen by blood serum-ELISA. According to ELISA milk test results, the lowest prevalance was detected as 1.92% in Aksu and the highest prevalance was detected as 34.55% in Yalvaç. It was concluded that there was a fair similarity (К= 0.214) between serum-ELISA and milk-ELISA, therefore, using milk serum in serological tests for BLV can not be considered as an alternative to using blood serum
