Fasulye Tohumlarındaki Viral Etmenlerin Saptanmasında Tanı Yöntemlerinin Duyarlılıklarının İncelenmesi
The Studies on Sensitivity of The Detection Methods of Viral Agents in Bean Seed Samples
The
study was conducted to find out the presence of viral infections in 70
bean seed samples providing from different seed companies and Ege University
Faculty of Agriculture Department of Horticulture. In order to determine the
sensitivity of the methods used in the detection of viral infections in bean
seed samples, the studies carried out in 2012 and 2013 years. To detect the
viral infections in bean seed samples; symptomatology, growing-on test,
biological test (mechanical inoculation), serological and molecular methods
were used. The presence of 5 viral agents in bean seed samples was checked by
RT-PCR, while the diagnosis of other 4 viruses was done with DAS-ELISA.
Considering the results obtained, the sensitivity of the methods used in this
work was investigated for the detection of viruses in bean seed samples. Out of
70 bean seed samples, the infection of certain viruses was detected in 69
samples by RT-PCR, 62 by DAS-ELISA, 48 by viral examinations and 30 by
biological indexing. In growing-on assays, the viral infections were found in
48 of 64 plants developing from 70 seed samples. BCMV, BCMNV, CMV,
and SMV were detected in seed samples, while AMV was not found. According to the
findings from this study, it was found that RT-PCR was the most sensitive
methods in the detection of the viruses in bean seed samples and the majority
of bean seed samples was infected with BCMV.
