Gen transferi-transfeksiyon Prostat kanseri STAMP2 stabil hücre hattının oluşturulması

Amaç: DNA- gen transferi, kanserde yapılan gen ekspresyon araştırmalarında anahtar rol oynamaktadır. Prostat kanserinin erken döneminde eksprese olan prostat spesifik genleri aradığımız bir projede altı transmembran domaini bulunan bir gen ailesini (six transmembrane protein of prostate- STAMP1 ve 2’yi) tanımladık. Gereç ve yöntem: PC3 hücre kültüründe (prostat kanseri kemik metastazı- STAMP gen ekspresyonları negatif), ektopik ekspresyon vektörleri kullanılarak STAMP üyelerinin ekspresyonu yerine konulmuş ve hücrelerdeki lokalizasyonları immünofloresan tekniği ile takip edilmiş ve doğrulanmıştır. Bulgular: HisMax-STAMP2’nin stabil hücre hatları oluşturulmuş ve stabilitesi doğrulanmıştır. Sonuç: Projenin belirtilen dizaynı ile ilgilenilen olası yolakları, çeşitli indüksiyonlar ve takip eden RT-PCR deneyleri ile incelemek mümkün olabilecektir.

Gene transfer-transfection establishment of prostate cancer STAMP2 stable cell line

Objective: DNA- gene transfer plays key role at gene expression studies of cancer. In a project that prostate-specific genes expressed in early term of prostate carcinoma is searched, a gene family with six transmembrane domain protein (six transmembrane protein of prostate- STAMP1 and 2) is identified. Material and method: In PC3 cell line (prostate cancer bone metastasis- STAMP gene expressions negative) ectopic expression vectors of STAMP members are placed in and localization is followed by immunofluoresence and confirmed. Results: Stable cell line of HisMax-STAMP2 is established and stability is confirmed. Conclusion: With the given design of the project, it will be available to search the interested pathways by the various inductions and following RT-PCR assays.
