Denizli horozu genital sistemi üzerinde histokimyasal araştırmalar

Bu çalışmada, Denizli horozu genital sisteminin ışık mikroskobik yapısı histokimyasal yöntemlerle araştırılmıştır. Ductuli efferentes’in lamina epitelyalisini çekirdekleri apikalde yer alan silyumlu hücreler ve çekirdekleri bazalde yer alan silyumsuz uzun hücrelerden oluşan tek katlı prizmatik epitel hücrelerinin meydana getirdiği görüldü. Ductus epididymidis epiteli ise apikallerinde büyük salgı granülleri içeren yalancı çok katlı prizmatik epitel yapısında ve lümeninde olgun spermatozoa yer alan bir kanaldı. Spermatogenik germ hücreleri, özellikle spermatogoniumlar bazik bir boya olan toluidin blue ile yoğun boyanmıştır. Fosfotungustik asit-hematoksilen boyama ile spermatogenik germ hücrelerinde, intersitisyel alandaki fibrillerde, bazal membranda ve kılcal damar çevresinde pozitif boyanma reaksiyonu gözlendi. Spermatogenik germ hücrelerinin çekirdek yapıları ve intersitisyel alandaki arteriolün iç elastik membranı orsein ile pozitif boyanma reaksiyonu göstermiştir. Sonuç olarak, Denizli horozu genital sisteminin yapısının histokimyasal olarak araştırılması, ırkın yapısal özelliklerinin tanıtımı bakımından önemli olabilir.

Histochemical investigations on the reproductive system in Denizli cock

In this study, the light microscopic structure of the reproductive system of Denizli cock was investigated by histochemical methods. The lamina epithelialis in the ductuli efferentes contains long ciliated and nonciliated simple cylindrical cells having apically and basally located nuclei, respectively. The ductus epididymidis is made of pseudostratified cylindrical epithelial cells which contain large secretory vacuoles in their apical portions, its lumen contains mature spermatozoa. Spermatogenic germ cells, especially spermatogoniums showed strong staining reactions with basic dye, toluidine blue. Positive staining reactions were observed in spermatogenic germ cells, fibers in interstitial region, basement membrane and around the capillary by phosphotungstic acid-hematoxylene staining. The nuclear structures of spermato-genic germ cells and the internal elastic membrane of arteriole in interstitial region showed positive orcein staining reaction. As a result, histochemical investigations of genital system structure of Denizli cock can be important in terms of the introduction of structural features of the race.
