Ratlarda oluşturulan deneysel artritin intraartiküler olarak verilen hyaluronik asitle tedavisi

Erken dönem dejeneratif osteoartrit tedavisi için henüz tam bir tedavi protokolü belirlenememiştir. Bu nedenle intraartiküler olarak verilen Hyaluronik Asit'in (HA) dejeneratif artrit tedavisinde ne derece etkili olduğunu araştırmak amacıyla 40 adet adult albino Wistar erkek ratın sağ arka diz eklemlerinde deneysel artrit oluşturuldu. Deneysel artrit eklem içine 0/2ml %1'lik formalin verilerek elde edildi ve ilaç verilmesinden 1 hafta sonra 10 rat öldürülerek artritin oluşturulduğu histopatolojik olarak ispatlandı. Diğer 30 rat 3 gruba ayrılarak birinci gruba intraartiküler 0.05 mi HA, ikinci gruba 0.05 mi serum fizyolojik verildi. Üçüncü gruba ise herhangi bir tedavi yapılmayarak kontrol grubu oluşturuldu. Haftada bir olmak üzere toplam üç kez uygulanan bu tedavilerin bitiminden 15 gün sonra öldürülen raflardan alınan örnekler, histopatolojik olarak modifiye Mankin sistemine göre değerlendirildi. Sonuçlar istatistiksel olarak Mann-Whitney U testi ile karşılaştırıldı, intraartiküler HA uyguladığımız deneklerde, serum fizyolojik uygulanan gruba ve kontrol grubuna kıyasla kıkırdakta daha fazla bir iyileşme (p

The treatment with intra-articular administration of hyaluronic acid on experimental arthritis in rats

A satisfactory protocol for the treatment of early stage degenerative osteoarthritis hasn't been determined yet. For this reason, in order to investigate how much Hyaluronic Acid (HA) administered intra-articularly is effective in degenerative arthritis treatment, experimental arthritis was formed in the rightjack knee joint of 40 adult albino Wistar male rats. The experimental arthritis was formed by administering 0.2 ml (%1) formalin into the joint and the formation of arthritis was demonstrated histopathologically by sacrificing 10 rats one week after the administration of formalin. The other 30 rats were divided into three groups and the first group was administered 0.05 ml HA intra-articularly. The second group was administered 0.05 ml serum physiologic intra-articularly. No treatment was applied for the third group and it was accounted as the control group. 15 days after termination of the treatment applied totally 3 times one per week, the samples obtained from the sacrificied rats were assessed according to modified Mankin system histopathologically. The resuts were compared with Mann-Whitney U test statistically. There were a better improvement in cartilage and a relative improvement in synovia in the group administered HA than those of the control group and serum physiologic group. As a result, it was concluded that HA is a clinically applicaple drug in the treatment of early stage degenerative osteoarhritis, but that it is necessary to perform comparative investigations with the other intra-articular treatments still in use.
