Doku ve Hücre Bütünlüğünün Korunması Açısından Üç Farklı Dekalsifikasyon Yönteminin Karşılaştırması
Kemik dekalsifikasyonu, kemik dokusunun histolojik incelenmesi için en önemli adımlardan biridir. Geleneksel yöntemlerde, dekalsifikasyon işlemi için farklı zayıf asitler uygulanır. Bu asitler doku içindeki hücresel yapıların organizasyonunu olumsuz etkilediğinden doku bütünlüğünü korumak için yeni yöntemler uygulanmaktadır. Bu çalışmada, kemik dekalsifikasyonunda yaygın olarak kullanılan %5 formik asit, asit içerikli RDO çözeltisi ve elektroliz dekalsifikatör sistemi EDS olmak üzere üç farklı kemik dekalsifikasyon yöntemini karşılaştırmayı amaçladık. Bulgularımız formik asit ile dekalsifikasyon sonucu doku ve hücre düzeyinde histomorfolojik ve immünohistokimyasal mikroskobik veri kaybının daha fazla olduğunu göstermiştir. Bununla birlikte, RDO ve EDS yöntemleri ile dekalsifiye edilen kemik kesitlerinde, HE ve trikrom boyamalarda ve immünreaktivite açısından histomorfolojik ve immünohistokimyasal verilerin daha kaliteli olduğu saptanmıştır. Bu sonuçlar, kemik dekalsifikasyonunda uzun süreli asit inkübasyonunun doku bütünlüğünü, hücre içi yapıları ve antijenik molekülleri bozarak görüntü kalitesini düşürdüğünü göstermektedir
Comparison of Three Different Decalcification Methods in Terms of Preservation of Tissue and Cell Integrity
Bone decalcification is one of the most important steps for histological examination of bone tissue. In conventional methods, different weak acids are applied for decalcification. As these acids adversely affect the organization of cellular structures within the tissue, new methods are applied to preserve tissue integrity. In this study, we aimed to compare three different bone decalcification methods, 5% formic acid, acid-containing RDO solution and electrolysis decalcifier system EDS , which are widely used in bone decalcification. Our findings showed that decalcification with formic acid resulted in more loss of histomorphological and immunohistochemical microscopic data at the tissue and cell level. However, histomorphologic and immunohistochemical data were found to be better in RDO and EDS groups. These results demonstrate that long-term acid incubation during bone decalcification degrades tissue integrity, intracellular structures, and degrades antigenic molecules
