İdrar N-asetil-beta-D-glukosaminidaz aktivitesi: İki farklı tayin yönteminin karşılaştırılması
Amaç: Lizozomal bir enzim olan N-asetil-beta-D-glukozaminidaz (NAG), renal tübüler hasarın er- ken belirleyicilerinden biridir. Bu çalışmada NAG aktivitesinin ölçümünde kullanılabilecek güvenlive kolayca uygulanabilir bir yöntemin belirlenmesi için p nitrophenyl N acetyl-B-D-glucosaminide(PNP-NAG) ve m, cresolsulfon phtaleinyl N, B, D glucosaminide (MCP-NAG) gibi kolorimetrik de- ğerlendirme imkanı veren iki farklı substrat kullanılarak yöntemlerin karşılaştırılması amaçlanmıştır.Yöntem: Bu çalışmada 40 sağlıklı ve 40 böbrek yetmezliği olan hasta idrarındaki NAG aktiviteleriPNP-NAG ve MCP-NAG substratları ile ölçülerek, sonuçlar karşılaştırıldı. PNP NAG substratı ile çalışır- ken örnekler jel filtrasyonundan önce ve sonra reaksiyona sokularak jel filtrasyonunun sonuçları neşekilde etkilediği test edildi.Bulgular: PNP-NAG (jel +), PNP-NAG (jel -) ve MCP-NAG yöntemleri ile elde edilen sonuçlar orta- lama olarak sırasıyla, hasta grubunda sırasıyla (27.5 U/L, 25.96 U/L ve 16.5 U/L) kontrol grubunasırasıyla (5.66 U/L, 5.38U/L ve 3.62 U/L) olup; değerler hasta grubunda kontrol grubuna göre yüksekbulunmuştur (p
Comperative study of determination of urinary N-acetyl-beta-D-glucosaminidase activity
Aim: One of lyzozomal enzymes N-Acetyl-Beta-D-glucosaminidase (NAG), is a sensitive parameterof renal tubular injury. In order to to determine a confidential and easily applicable method we haveplan to measure and compare the results of NAG activities with two different substrates namelyp-nitrophenyl-N-acetyl-B-D-glucosaminide (PNP-NAG) and m-cresolsulfon-phtaleinyl-N-B-D-glu- cosaminide (MCP-NAG) .Material-Methods: We measured urinary NAG levels using PNP-NAG and MCP-NAG substrates in40 healty persons, and in 40 renal failure patients.
