Herbisit direncli transgenik tütün bitkilerinin geliştirilmesi
Bu çalışmada aktif maddesi glufosinat amonyum olan herbisitlere dirençli transgenik tütün bitkilerinin geliştirilmesi amacı ile yapılan araştırmamaların ilk sonuçlan sunulmaktadır. İzlenen strateji, herbisitin aktif maddesi olan glufosinat amonyum'un detoksifıkasyonunu olası kılan bir enzim olan fosfonitrisin asetil transferaz'ın (PAT) sentezinden sorumlu BAR geninin tütün bitkilerine transferi şeklindedir. Gen transferi Agrobacterium tumefaciens LBA4404 suşları kullanılarak, "binary" vektör stratejisi ile gerçekleştirilmiştir. Gen transferi sonrası, kontol bitkiler için letal olan 4 mgllitre derişimde glufosinat amonyum varlığında yaprak disklerinde kallus ve gövde oluşumu gözlenmiştir.
Development of herbicide resistant transgenic tobacco plants
This work describes the preliminary results of our resarch towards development of transgenic tobacco plants resistant against herbicides with glufosinate ammonium as the active ingradient. The strategy was to transfer BAR gene to tobacco plants, which codes for phosphinotricin acetyl transferase (PAT), an enzyme enabling the detoxification of the herbicide's active ingredient glufosinate amonium. Gen transfer was performed by using Agrobacterium tumefaciens LBA4404 strains according to binary vector strategy. After gene transfer, callus and shoot generation was observed in the presence of 4 mg/liter concentration of glufosinate ammonium, which was lethal to control plants.
