Malignant Meme Tümörlü Köpeklerde Plazma IGF-2 Konsantrasyonu ve Tümör Dokusundaki Ekspresyonu

Bu çalışmanın amacı, malignant meme tümörlü köpeklerde insülin benzeri büyüme faktörü-2 (IGF-2)'nin plazma konsantrasyonu ve tümör dokusundaki ekspresyonunu değerlendirmektir.  Çalışmada toplam 15 köpek kullanıldı. Sekiz meme tümörlü köpek grup 1'i, 7 sağlıklı köpek ise grup 2'yi oluşturdu. Köpeklerin diöstrus döneminde oldukları vaginal sitolojik muayene ile belirlendi. Abdominal organların ultrasonografik muayenesi ve toraksın radyografik bakısı yapılarak metastaz değerlendirilmesi yapıldı. Köpeklerden kan örnekleri alındı, plazmaları elde edildi ve IGF-2 konsantrasyonu ELISA yöntemi ile belirlendi. Grup 1'de, tümörler “Dünya Sağlık Örgütü” (WHO)'nun belirlediği “Klinik Derecelendirme Sistemi”ne göre sınıflandırıldı ve komple unilateral mastektomi tekniğiyle uzaklaştırıldı. Tümörlerin histolojik olarak malignite derecesi Nottingham yöntemine göre tubul formasyonu, nüklear pleomorfizm ve mitotik hücre sayısına bağlı olarak değerlendirildi. Histopatolojik incelemelerde 4 karsinosarkom, 1 intraduktal papillar karsinom, 1 karsinom, 1 kompleks tip karsinom ve 1 miks tip karsinom tanısı konuldu. İmmunohistokimyasal incelemede tümör dokusunda meme asinus epitel hücreleri, miyoepitel hücreleri ve bazı damar düz kas hücrelerinde IGF-2 ekspresyonunun olduğu, duktus epitel hücrelerinde ise olmadığı gözlendi. Sağlıklı köpeklere göre, malignant meme tümörlü köpeklerde plazma IGF-2 konsantrasyonundaki artış istatistiksel olarak anlamlı bulundu (P˂0.05).
