$[D-Ser(tBu)^6, Pro9-Net^]$ -LHRHa ve metoklopramidin farklı kombinasyonlarda kullanımı ile sazan (Cyprinus carpio) anaçlarında yumurtlamanın uyarılması

Bu çalışmada sazan anaçlarında yumurtlamayı uyarmak amacıyla, olgun anaçlara $[D-Ser(tBu)^6,Pro^9-NEt]$ -LHRHa (LHRH-a) yüksek dozda tek başına (50 ug/kg) ve sabit dozda (20 pg/kg), bir dopamin reseptör antagonisti olan Metoklopramit'in (MET) farklı dozları ile birlikte (5, 10, 20, 40 mg/kg) tek enjeksiyonda uygulanarak, lokal şartlar altında etkileri belirlenmiş ve 24 °C su sıcaklığında klasik hipofız ekstraktı uygulaması (çift enjeksiyon) ile karşılaştırılmıştır. Kontrol grubuna ise sadece fizyolojik tuzlu su (% 0,7 NaCl ) verilmiştir. Kontrol grubunda ve LHRH-a'nın MET'in düşük dozları (5, 10 mg/kg) ile birlikte verildiği gruplarda yumurtlama gözlenmezken, LHRH-a'nın yüksek dozda'tek başına verildiği grupta (50 pg/kg) yumurtlama oranı (YO) düşük gerçekleşmiştir. LHRH-a'nın MET'in yüksek dozları (20, 40 mg/kg) ile birlikte verildiği gruplarda ve hipofız enjekte edilen gruplarda YO yüksek olarak gerçekleşmiştir. LHRH-a+MET verilen gruplarda enjeksiyondan sonra yumurtlama süresi (YS), hipofız ekstraktı verilen gruba göre yaklaşık 2 kat daha kısalmıştır (sırasıyla 14-16 saat, 24-26 saat). Yumurtlayan gruplar arasında anaçların yumurtlama indeksi (Yİ) ve yumurtaların döllenme ve açılma oranları (DO, AO) açısından farklılık gözlenmemiştir. Sonuç olarak, 20 ug/kg LHRH-a+20 mg/kg MET uygulaması ile sazan anaçlarında yumurtlama, tek enjeksiyonda ve hipofiz enjeksiyonuna kıyasla daha kısa bir süre içinde ve yumurta kalitesinde düşmeye neden olmaksızın başarılı bir şekilde uyanlabilmiştir. Bu kombinasyonun uygulanması, sazan yetiştiriciliğinde daha etkin bir anaç ve kuluçkahane yönetimi açısından önemli faydalar sağlayabilir.

Spawning induction in the common carp (Cyprinus carpio) using different combinations of [D-Ser(tBu)^6,Pro^9-Net] -LHRHa and metoclopramide

The effects of a single administration of $[D-Ser(tBu)^6,Pro^9-NEt]$-LHRHa at high dose alone (50 yg/kg), and at constant dose (20 pg/kg) combined with different doses of the dopamine receptor antagonist Metoclopramide (5,10, 20,40 mg/kg) for the induction of spawning in common carp broodstocks were determined under local hatchery conditions and compared with classic carp pituitary extract (CPE, double injection) application (water temperature 24 °C). Fish injected with physiological saline (0.7% NaCl) were used as a control group. No spawning was observed in the control group or in the groups given LHRH-a combined with low (5,10 mg/kg) doses of MET. The spawning ratio was low in the group given a high dose of LHRH-a alone (50 ug/kg), while it was high in the groups given LHRH-a combined with high (20, 40 mg/kg) doses of MET, and in the group injected with CPE. Latency period in the LHRH-a+MET groups was approximately half of that in the CPE group (14-16 and 24-26 h, respectively). There were no differences between spawned groups with respect to the spawning index of broodstocks, or the fertilization/hatching rate of eggs. It was concluded that spawning can be induced successfully in common carp broodstocks with 20 pg/kg LHRH-a+20 mg/kg MET, jn a.single injection, in a shorter latency period and without a negative effect on egg quality, compared to CPE treatment. This combination may be useful for more effective broodstock and hatchery management in common carp culture.
