Üç farklı kuluçkahanedeki damızlık gökkuşağı alabalığı (Oncorhynchus mykiss Walbaum, 1792) stokları arasında genetik çeşitliliğin RAPD-PCR yöntemiyle belirlenmesi üzerine bir araştırma

Bu araştırmada RAPD-PCR yöntemi kullanılarak, Uç farklı kuluçkahanedeki damızlık gökkuşağı alabalıkları (Oncorhynchus mykiss) arasında genetik çeşitliliğin belirlenmesi amaçlanmıştır. Muğla-Fethiye, Burdur-Gölhisar ve Denizli-Çameli yöresinde üretim yapan üç işletmeye ait toplam 45 damızlık balık materyal olarak kullanılmıştır. Üç populasyonda hesaplanan genetik benzerlik değerleri oldukça düşük bulunmuştur. Fethiye populasyonu içinde ortalama 0.429, Çameli populasyonu içinde 0.311 ve Gölhisar populasyounda 0.348 olarak hesaplanmıştır. Ortalama populasyon içi genetik farklılık değerleri ise Fethiye populasyonunda 0.571, Çameli populasyonunda 0.689 ve Gölhisar populasyonunda 0.652 olarak hesaplanmıştır. Populasyonlar arası genetik farklılık değeri olduça yüksek bulumuştur. En yüksek genetik farklılık değeri Fethiye-Gölhisar populasyonları arasında 0.752 olarak, en düşük ise Fethiye-Çameli populasyonları arasında 0.659 olarak belirlenmiştir.

A study on the determination of genetic variability between rainbow trout (Oncorhynchus mykiss Walbaum, 1792) broodstocks reared in three different hatcheries by RAPD-PCR method

In this study, genetic variability between and whithin rainbow trout broodstocks rared in three different hatcheries was investigated by RAPD-PCR method. For this purpose, total 45 fish used from three trout farm located in Muğla-Fethiye, Burdur-Gölhisar and Denizli- Çameli region. The genetic similarities calculated for three population were found quite low. The mean genetic similarity was found as 0.429 within Fethiye population, 0.311 for Golhisar population and 0.348 for Çameli population. Genetic diffirences were calculated within population, 0.571 in Fethiye, 0.689 in Çameli and 0.652 in Gölhisar. Genetic variation between populations was found very high. The highest variation was determinated as 752 between Fethiye and Gölhisar populations and the least genetic variation was calculated as 0.659 between Çameli and Fethiye populations.
