The use of X- Radiography technique for the determination of gastric evacuation rate of raainbow trout ( Oncorhynchus mykiss Walbaum,1972 )
The use of X-Radiography technique for the determination of gastric evacuation rate of rainbow trout (Oncorhynchus mykiss Walbaum, 1792). Eighty female rainbow trout (O. mykiss) (205.2±5.6 g) were fed a test diet containing radio opaque glassbeads "ballotini" (0.6-0.9 mm diameter) and 10 fish were sacrificed at different time intervals (0, 6, 12, 18, 24, 30, 36 and 48 hours following feeding) and X-rayed. The amount of feed remaining in the stomach for each time interval was determined following the quantification of the number of glass beads in the developed X-ray sheets and the gastric emptying rate of trout was modelled. In order to test the efficiency of X-radiography technique, the stomach contents of X-rayed fish were dried at 105°C for 24 hours and the true gastric evacuation model was produced by dry matter quantification. Finally, two emptying curves generated from both X-radiography and serial slaughter techniques were statistically compared by regression analysis. It was observed that the evacuation curves obtained by X-ray and serial slaughter methods were significantly different (p<0.05). This was because radio-opaque" glassbeads did not demonstrate a typical evacuation pattern and selectively retained in the cardiac stomach region of trout. It was suggested that the X-ray technique for the determination of stomach evacuation of rainbow trout is not valid.
