Leishmanialar memeli konakta makrofaj hücreleri içinde amastigot, vektörde ise promastigot olarak yaşayan bir doku parazitidir. Propolis, balarıları tarafından bitkilerden toplanan ve mumla karıştırılarak kovan içerisinde birçok amaca yönelik olarak kullanılan doğal bir üründür. Bu çalışmanın amacı, Leishmania tropica ile infekte insan makrofaj hücrelerinde interlökin (IL)-1β, IL6 ve IL-12 sitokin seviyelerine propolisin etkilerini araştırmaktır. Çalışma; makrofaj (M), makrofaj+parazit (M+P) ve makrofaj+parazit+propolis (M+P+Pr) olmak üzere üç farklı grup üzerinde yapıldı. U-937 insan makrofaj hücreleri, % 10 fetal bovin serum ve % 2 penisilinstreptomisin ilaveli RPMI-1640 besiyerinde üretilerek, son konsantrasyonu 1×106 hücre/ ml olacak şekilde 24’lük hücre kültür plaklarına dağıtıldı. L.tropica promastigotları, makrofaj:promastigot oranı 1:10 olacak şekilde makrofaj hücreleri üzerine ilave edildi. Makrofajlar içine giren promastigotlar, amastigot forma geçti. Son konsantrasyonu 64 µg/ml olacak şekilde propolis ilave edildi. Plaklar %5 CO2’li ortamda 37oC’de 24 saat inkübe edildi ve inkübasyon sonunda üç ayrı grubun süpernatanlarında IL-1β, IL-6 ve IL-12 sitokin seviyeleri ELISA yöntemiyle ölçüldü. IL-12 sitokin seviyesi M+P+Pr grubunda diğer iki gruba göre daha yüksek bulundu (p<0.05). IL-1β ve IL-6 sitokin seviyeleri propolis uygulanan grupta M+P grubuna göre daha düşük bulundu (p<0.05). Bu çalışma, propolisin L.tropica ile infekte makrofaj hücrelerinde IL-1β ve IL-6 sitokinlerinin baskılanmasına ve IL-12 seviyesinin artmasına sebep olduğunu ve böylece immün yanıtı desteklediğini göstermiştir

Leishmania, a tissue parasite, live in macrophage cells in the mammalian host as amastigote, in vector as promastigote. Propolis, collected from plants by bees and hives in mixed wax is a natural product used for many purposes. The aim of this study was to investigate the effects of propolis on the levels of interleukin (IL)-1β, IL-6 and IL-12 cytokines on Leishmania tropica infected human macrophage cells. This study was performed on three different groups of macrophage (M), macrophage+parasite (M+P), and macrophage+parasite+propolis (M+P+Pr). U-937 human macrophage cells were grown in RPMI-1640 supplemented with 10% fetal bovine serum and 2 % penicillin-streptomycin. The final cell concentration as 1×106cells/ml were dispended in 24-well cell culture plates. L.tropica promastigotes were added by a ratio of 1 macrophage cell:10 promastigotes to be on the macrophage cells. Promastigotes were entered into macrophage cells and transformed into amastigotes form. The final concentration of propolis is 64 µg/ml were added. The plates were incubated at 37oC and in 5% CO2 incubator for 24 hours. At the end of incubation, IL1β, IL-6 and IL-12 cytokine levels were measured in supernatants of the three different groups by ELISA. IL-12 cytokine levels in the M+P+Pr group were found higher than the others groups. (p<0.05). IL-1β and IL-6 cytokine levels in the M+P+Pr group were found lower than in the M+P group (p<0.05). This study shows that, propolis causes the supression of IL-1β and IL-6 cytokines and the increases IL-12 cytokine levels on the L.tropica infected macrophage cells and this also supports the immun response
