FAREDE AKUT AĞRI MODELİNDE SERTRALİNİN OLUŞTURDUĞU ANTİNOSİSEPTİF CEVABA KAFEİNİN ETKİSİ

Antidepresan ilaçlar kronik ağrı durumlarının tedavisinde oldukça yaygın kullanılırlar. Bir antidepresan ilaç olan sertralinin de deneysel ve klinik çalışmalarda çeşitli ağrı durumlarında etkili olduğu gösterilmiştir. Ancak sıkça tüketilen bir alkaloid olan ve bazı antidepresanların antinosiseptif etkilerini değiştirdiği gösterilen kafeinin, sertralinin oluşturduğu antinosisepsiyon üzerine herhangi bir etkisinin olup olmadığına yönelik bir bilgi bulunmamaktadır. Bu nedenle; sunulan çalışmada akut olarak farelere uygulanan sertralinin oluşturduğu antinosisieptif etki üzerine kafeinin olası rolünün araştırılması amaçlanmıştır. Çalışmada 32 adet erkek ve dişi Swiss albino fare; SF+SF, kafein (5 mg/kg)+SF, SF+sertralin (40 mg/kg) ve kafein (5 mg/kg)+sertralin (40 mg/kg) injeksiyonu uygulanan dört gruba ayrılıp uygulanan drog kombinasyonlarının etkisi hot-plate analjezimetresi ile test edildi. Sertralinin 30., 45. dakikalarda antinosiseptif etki gösterdiği ve bu etkinin 60. dakikada azaldığı, kafeinin ise tek başına herhangi bir antinosiseptif etki göstermediği, sertralin ve kafein kombine edildiğinde sertralinin antinosiseptif etkisinde istatistiksel olarak anlamlı bir değişiklik meydana gelmediği belirlendi. Sonuç olarak farede sertralinin akut uygulama ile oluşturduğu antinosiseptif cevabın uygulanan dozda kafein ile değişmediği gösterildi

Effect of Caffeine on Antinociceptive Response of Sertraline in Acute Pain Model in Mice

Antidepressant drugs are widely used in the treatment of chronic pain states. The effectiveness of sertraline, an antidepressant drug, has been shown in different pain states in experimental and clinical studies. However, there is no information whether caffeine, a widely consumed alkaloid known to change the antinociceptive effects of some antidepressant drugs, has an effect on sertraline induced antinociception. Therefore, in the present study, we aimed to investigate the possible role of caffeine on sertraline induced acute antinociceptive response in mice. In the study, 32 male and female Swiss albino mice were separated into four groups (n=8) and injected with saline+saline, caffeine (5mg/kg)+saline, saline+sertraline (40mg/kg) and caffeine (5mg/kg)+sertraline (40mg/kg). The effects of these drug combinations were tested with a hot-plate analgesiameter. It was found that sertraline induced an antinociceptive effect at the 30th and 45th minutes, and the effect decreased at the 60th minute, whereas caffeine alone did not induce an antinociceptive response. When sertraline was combined with caffeine, the antinociceptive effect of sertraline did not change significantly. As a result, it was deduced that the antinociceptive response of sertraline in acute application in mice was not affectedby an applied dose of caffeine
