DİAZİNON’UN SIÇAN TESTİSİ ÜZERİNE ETKİLERİNİN HİSTOKİMYASAL OLARAK İNCELENMESİ

Bu çalışmada, Kayseri bölgesinde tarım alanlarında kullanılan insektisitlerden biri olan diazinonun histokimyasal olarak araştırılması amaçlanmıştır. Araştırmada kullanılan sıçanlar 5 gruba ayrıldı. Birinci deney grubuna 1 gün LD50 dozunun (1250 mg/kg) 1/10’u, ikinci deney grubuna 2 gün LD50 dozunun 1/20’si, üçüncü deney grubuna 4 gün LD50 dozunun 1/40’ı, dördüncü deney grubuna 8 gün diazinon’un LD50 dozunun 1/80’i serum fizyolojikle eritilerek sıçanların ağırlıklarına göre 0.75-1 ml/kg/gün gavajla oral olarak verildi. Beşinci grup sıçanlar kontrol grubu olarak değerlendirildi. Işık mikroskobunda histolojik inceleme için rutin takip yöntemleri uygulandıktan sonra elde edilen testis dokusu kesitlerinde her grup için ayrı ayrı Johnsen testiküler biyopsi skoru hesaplandı, seminifer tübül çapları ölçüldü ve histolojik olarak değerlendirildi. inceleme için TUNEL metodu kullanıldı ve germ hücre apoptozisi değerlendirildi. STÇ’ları ve Apoptotik hücre oranları incelendiğinde gruplar arasında anlamlı farklılık saptandı (p<0.05). İkinci deney grubunun apoptotik hücre ortalaması diğerlerine göre daha yüksek bulundu. JTBS’ları incelendiğinde, gruplar arasında anlamlı farklılık saptandı (p<0.05). Kontrol grubu ile bütün gruplar arasındaki farklılık anlamlı bulundu. Diazinonun LD50 dozunun 1/10, 1/20, 1/40 ve 1/80’inin uygulandığı sıçanların testis dokularında spermatojenik hücrelerin, diazinondan değişik şekillerde etkilendiği, çoğalan engelleyerek infertiliteye neden olabileceği sonucuna varıldı

Histochemical Investigation of Effects of Diazinon on the Rat Testes

The aim of this study is to elucidate the testicular histochemical effects of diazinon, which is an insectiside used for agricultural purposes in areas Kayseri Region. The rats used in the study were divided into 5 groups.To the first experimental group for one day LD50 dose of (1250 mg/kg) 1/10, to the second experimental group for two days the LD50 dose of 1/20, to the third experimental group for four days the LD50 dose of 1/40 , to the forth experimental group for eight days LD50 dose of 1/80 diazinon were given with 0.75-1 ml/kg /day gavage orally after melted in the serum physiologic according to the weights of rats. The rats in the fifth group were asigned to be the control group. After the routine tracking methods were applied for the histologic analysis in the light microscobe, in the obtained testes tissues fractions Johnsen testicular biopsy score seminiferous tubule diameters were measured and evaluated histologically. For the analysis in the immunofloresance microscobe, TUNEL method was used and apoptosis of germ cell was evaluated. When the groups were analysed according to their STÇS and apoptotic cell average, a significant difference was observed between the groups (p<0.05). When analysed in terms of the JTBS there was a significant difference between the groups (p<0.05). The difference between the control group and the other groups was also significant.spermatogenic cells in the testes tissues, treated with 1/10, 1/20, 1/40, 1/80 LD50 dose diazinon, were affected in different ways. The proliferation of apoptotic cells caused infertility by hindering spermatogenesis
