Develi yöresinde sığır brusellozunun serolojik testlerle (RBPT, SAT, C-ELISA, CFT) teşhisi
Özet: Bu çalışma, Kayseri ili Develi ilçesinde abortyapan sığırların serumlarında 4 farklı serolojik testkullanılarak Brucella abortus karşı şekillenen antikorlarınortaya konulması; kullanılan serolojik testlerin teşhistekiduyarlılıklarının saptanması ve bölgede sığır brusellozungörülme oranının tespit edilmesi amacıylagerçekleştirilmiştir. Develi ilçesinde abort yapan 200 baş sığırdan kanörnekleri toplanmıştır. Serumlar Rose Bengal Plate Test(RBPT)’i, Serum Aglütinasyon Test (SAT)’i, KompetatifELISA (C-ELISA) ve Komplement Fikzasyon Test(CFT)’ine tabi tutulmuş, testlerin duyarlılıklarınındeğerlendirilmesinde CFT standart test olarak kabuledilmiştir. İncelenen 200 serumunun 57 (% 28,5)’si RBPT ile pozitifbulunurken geri kalanın negatif olduğu saptandı. Serumaglütinasyon test ile serumların 22 (% 11)’sinin pozitif,168 (% 84)’inin negatif ve 10 (% 5)’unun şüpheli olduğubelirlenirken C-ELISA ile 36 (% 18)’sı pozitif ve 164 (%82)’ü negatif olarak tespit edildi. Diğer taraftan CFT ile30 (% 15)’u pozitif, 163 (% 81,5)’ü negatif ve 7 (% 3,5)’sişüpheli olarak değerlendirildi. Brusellozun teşhisinde herüç testinde duyarlı oldukları, CFT ile C-ELISA arasındadiğer testlere göre daha iyi bir uyumluluğun bulunduğusaptandı. Develi ilçesinde atık yapan sığırlardabrusellozis görülme oranının % 15 olduğu belirlendi. Sonuç olarak, Develi yöresinde sığırlarda atık olgularınınyüksek bir görülme oranına sahip olduğu ve halk sağlığıaçısından da büyük bir risk oluşturabileceğibelirlenmiştir. Kısa sürede ve güvenilir sonuçlaralındığından sığır brusellozunun serolojik teşhisinde C- ELISA’nın daha üstün olduğu kanaatine varılmıştır.
The diagnosis of bovine brucellosis with serologic tests (RBPT, SAT, C-ELISA, CFT) in Develi district
Summary: This study was carried out to detect theantibodies against Brucella abortus in the sera of abortedcows reared in Develi district of Kayseri province withfour different serological tests and to compare thesensitivity of the applied serological tests as well as todetermine the incidence of bovine brucellosis in theregion. Blood samples were collected from the 200aborted cows in Develi district. Sera were tested withRose Bengal Plate Test (RBPT), Serum AgglutinationTest (SAT), Competetive ELISA (C-ELISA) andComplement Fixation Test (CFT) and the CFT wasaccepted as the standard test for the evaluation of thesensitivity of the tests. Of 57 (28.50 %) out of 200 testedsera were found to be positive with RBPT and theremaining sera were found as negative. Of 22 sera (11 %)were found to be positive, 168 (84 %) of the sera werefound to be negative and 10 of the sera were found to besuspicious with SAT whereas 36 (18 %) of the sera werepositive and 164 (82 %) of the sera were negative with C- ELISA. On the other hand, CFT revealed that 30 (15 %)of the sera were positive, 163 (81.50 %) of the sera werenegative and 7 (3.50 %) of the sera were suspicious. Forthe diagnosis of brucellosis, all of the tests were found tobe sensitive, the results of C-ELISA were found to bemore consistent with the results of CFT than the othertests. The frequency of brucellosis in aborted cows inDeveli district was determined as 15 % according to theresults of the tests. In conclusion, it has been determinedthat the incidence of abortus in cattle in Develi district ishigh, and it may pose a high risk for public health.Because rapid and reliable results were obtained with theC-ELISA, this test is considered to be superior to otherinvestigated tests for the serological diagnosis of bovinebrucellosis.
