Böceklerde Farklı Muhafaza Koşulları ve Zamanın Mitokondrial DNA Kalitesi Üzerine Etkisi: Bir Diptera (Tephiritidae) Örneği Çalışması

Bu çalışmada geleneksel öldürme yöntemleri ile elde edilmiş ve farklıkoşullarda muhafaza edilen Tephritidae familyasına ait Euarestabullans (Wiedemann, 1830) örneklerinden DNA izole edilerekmuhafaza koşulları ve muhafaza sürelerine bağlı olarak DNA kalitesive PZR başarıları araştırılmıştır. Bu anlamda standart barkod geniolan mitokondriyal sitokrom oksidaz I (COI) gen bölgesikullanılmıştır. 15 yıllık örneklerden kuru ortamda (müze materyali)muhafaza edilenlerin yaşı ile PZR başarısı arasında tam bir ilişkigözlemlenmemiştir. Ancak alkolde (%95’lik etil alkol) saklananörneklerde ise DNA kalitesi ve PZR başarısı yüksek bulunmuştur.

The Effects of Different Storage Conditions and Storage Time on The Quality of The Mitochondrial DNA on Insects: Sample Study is A Fruit Fly (Diptera: Tephiritidae)

In this study, DNA was isolated from the samples of Euaresta bullans (Wiedemann, 1830) belonging to the Tephritidae family which was obtained by traditional killing methods and kept under different conditions. In this sense, the standard barcode gene mitochondrial cytochrome oxidase I gene (COI) was used. The lack of relationship observed between the age of samples that were kept in dry conditions (museum material) for 15 years and PCR success. However, the DNA quality and PCR success of the samples stored in alcohol (95% ethyl alcohol) were found to be high.
