Testiste Spermatogenik Hücrelerde 1 İntegrin ve Fibronektin Dağılımı

Amaç: Fibronektin β1 integrinle etkileşerekhücre adezyonu, göçü ve proliferasyonundagörev alır. Bu çalışmada gelişmekte olan testisdokusunda 1 integrin ve fibronektindağılımını immünohistokimyasal yöntemlegöstermek amaçlandı.Gereç ve yöntem: Çalışmada; Wistar cinsierkek sıçanlar kullanıldı. Sıçanlar, yaşlarınabağlı olarak ver her grupta 7 adet olmak üzereyenidoğan, 3, 10, 15, 21 günlük, 1, 2 aylık veergin olmak üzere 8 gruba ayrıldı. Dokularafibronektin ve 1 integrin içinimmünhistokimyasal yöntem uygulandı.Bulgular: Testiste fibronektinin bir bazalmembran ve bağ doku bileşeni olarakseminifer tüplerin bütünlüğüne katkıdabulunduğu ve hücre-hücre etkileşiminden çokhücre-bazal membran ve bazal membraninterstisyel doku etkileşiminde işlevsel olduğugörüldü. Fibronektin tutulumunun en belirginizlendiği alanların peritübüler alan, bazalmembran ve myoid hücre katmanı olduğugörüldü. 1 integrin tutulumunun, özelliklegelişimin ileri evrelerinde, peritübüler dokudave bazı spermatogenik seri hücrelerindebelirgin olduğu izlendi.Sonuç: β1 integrinin ise daha çok hücre-hücreetkileşiminde ve dolayısıyla spermatogenezisteetkin olabileceği kanısına varıldı.

Localization of 1 integrin and fibronectin on spermatogenic cells in rat testis

Aim: Fibronectin interacts with its receptor, 1integrin, and takes place in cell adhesion,migration and proliferation. In this study, weexamined the distribution of fibronectin and 1integrin of testis tissues in differentdevelopmental stages withimmunohistochemical methods.Material and methods: Wistar rats were usedin the study and divided into 8 groupsaccording to their ages: newborn, 3 days, 10days, 15 days, 21 days, 1 month, 2 months andadult. Testis tissues were examinedimmunohistochemically for fibronectin and 1integrin.Results: The best fibronectin reactionsoccurred in peritubular area, basementmembrane and myoid cell layer and thesereactions became more apparent in later stages.Besides we observed that there was no reactionin seminiferous tubule epithelium till adultstage. In adult rat testis tissue, fibronectinimmune staining was mild to weak in celllayers of seminiferous epithelium. 1 integrinimmune staining was diffusely strong inperitubular tissue and spermatogonia fromseminiferous epithelium. Reaction wasrestricted to cytoplasm and membranes of thegerminal cells.Conclusion: As a result, fibronectin in testis ismore functional in cell-basement membraneand basament membrane-interstitial tissueinteraction than cell-cell interaction; and 1integrin is essential in cell-cell interaction andspermatogenesis.
