Bu çalışmada, Harran Üniversitesi Veteriner Fakültesi’ne getirilen bir siyah kuğunun (Cygnus atratus) cinsiyetinin polimeraz zincir rekasiyonu (PZR) yöntemi ile belirlenmesi amaçlanmıştır. Bu amaçla CHD (chromodomain-helicase-DNAbinding) lokusunun hedef bölgesi iki farklı primer çifti (1237L-1272H ve P2-P8) yardımıyla PZR yöntemi ile çoğaltıldı ve PZR ürünleri agaroz jel elektorofrezi ile ayrımlandı. Her iki primer çifti ile elde edilen PZR ürünleri için agaroz jelde farklı uzunlukta iki bant gözlendi. Kuğunun dişi olduğu kanaatine varıldı.

The aim of the present study was to identify the gender of a black swan (Cygnus atratus) which was brought to Harran University Faculty of Veterinary Medicine by using polymerase chain reaction (PCR) technique. Target region of CHD (chromodomain-helicase-DNA-binding) locus was amplfied with PCR by using 1237L-1272H or P2-P8 primer pairs and PCR products were separated by using agarose gel electrophoresis. On agarose gel two bands were observed for both primer pairs. The results indicated that the swan was female.
