TİP II DİABETES MELLİTUS'LU HASTALARIN ERİTROSİT İÇİ Cu,ZnSOD, CAT ve GSH-Px ANTİOKSİDAN ENZİM DÜZEYLERİNİN ARAŞTIRILMASI

Bu araþtýrmada Tip II Diabetes Mellitus'lu hastalarýn eritrosit içi Süperoksit Dismutaz (Cu,Zn-SOD), Katalaz (CAT) ve Glutatyon Peroksidaz (GSH-Px) enzim aktiviteleri ölçüldü. Araþtýrma grubu, komplikasyonlarý bulunmayan Tip II Diyabetli 50 kiþi ve gönüllü saðlýklý 50 kiþiden oluþturuldu. Kontrol grubu eritrosit içi Cu,Zn-SOD, CAT ve GSH-Px aktiviteleri sýrasýyla 1071,7 ± 331,5 Ü/gHb, 175,4 ± 38,8 K/gHb, ve 1381,8 ± 690,6 Ü/gHb iken, Tip II Diabetes Mellitus'lu hastalarda ayný enzimlerin aktiviteleri sýrasýyla 653,2 ±215,5 Ü/gHb, 196,3 ±53,3 K/gHb, ve 1236 ±478,6 Ü/gHb bulundu. Kontrole göre hastalarýn SOD aktivitesi anlamlý þekilde düþmüþken, CAT aktivitesi ise anlamlý þekilde yüksek olarak tespit edildi (p<0,05). Kontrol ve hasta grubu GSH-Px enzim aktivitesi arasýnda anlamlý bir fark gözlenmedi. Bu verilerin ýþýðý altýnda, Tip II Diabetes Mellitus gibi glukoz toleransýnýn bozulduðu durumlarda serbest oksijen radikallerinin aþýrý üretilebileceði ve böylece eritrosit içi oksidan/antioksidan dengede bozulma olabileceði kanaatine varýldý.

THE INVESTIGATION OF ERYTHROCYTE Cu,Zn-SOD, CAT AND GSH-Px ANTIOXIDANT ENZYME LEVELS IN PATIENTS WITH TYPE II DIABETES MELLITUS

In this study, the activities of Superoxide Dismutase (Cu,Zn-SOD), Catalase (CAT) and Glutathione Peroxidase (GSH-Px) were measured in the erythrocyte of the patients with type II Diabetes Mellitus. The research group included 50 type II diabetic patients without complications and 50 healthy volunteers. The activities of erythrocyte enzymes Cu,Zn-SOD, CAT and GSH-Px from control group were determined as 1071.7 ± 331.5 U/gHb, 175.4 ± 38.8 K/gHb, and 1381. ± 690.6 Ü/gHb respectively. In the patient with type II Diabetes Mellitus activity of the same enzymes were 653.2 ±215.5 U/gHb, 196.3 ±53.3 K/gHb, 1236 ±478.6 U/gHb . While the SOD activity of patients was significantly lower, the CAT activity was determined to be significantly higher than that of control group (p<0.05). No significant difference between the GSH-Px enzyme activity of patients and control group was observed. Considering these results, we conclude that the conditions where the glucose tolerance impaired such as Type II Diabetes Mellitus may cause in excessive production of the oxygen radicals and the oxidant/antioxidant balance in erythrocytes may be disturbed.
