Programlı Hücre Ölümü; Literatür Bilgilerinin Türkçe Derlemesi
Programlı hücre ölümü, embriyogenez ve metamorfoz, sırasında istemeyen hücrelerin ve gelişimi endokrin bağımlı dokuların ortadan kaldırılmasındave normal doku homeostasisinin sağlanmasında önemlidir. Bu işlem enerji bağımlıdır ve ölümü gerçekleşecek olan hücrelerin kendi aktif katılımları ilegerçekleşmektedir. Programlı hücre ölümü, intrinsik ve ekstrinsik yolak olarak da adlandırılan mitokondriyal yolak ve ölüm reseptör yolağı olarak ikişekilde oluşmaktadır. Bunlar dışında, perforin/granzim yolağı ile gerçekleşen hücre ölümü de programlı hücre ölümü içerisinde incelenebilir. Bu her üçyolak, kaspaz3/7’nin aktifleştirilmesi ve programlı hücre ölümü sonrasında gerçekleşen kromatin yoğunlaşması, DNA parçalanması, nükleer vesitoplazmik proteinlerin parçalanması gibi olaylarda birleşmektedirler. Hücre dışı uyarılar ile başlatılan ekstrinsik yolak, hücrelerin bu uyaranlaraverdikleri cevaba bağlı olarak iki farklı şekilde işlemektedir. Bu yolakta, Tip I olarak adlandırılan hücreler DISC bileşimi içinde aktifleşen kaspaz-8’inkaspaz-3’ü aktifleştirmesine ihtiyaç duyarlar. Tip II hücrelerde ise, DISC bileşimi ve buna bağlı olarak da aktif kaspaz-8 oluşumu azalmıştır. Programlıhücre ölümünün gerçekleştirilebilmesi için, aktifleşen kaspaz-8 tarafından sitozolik BID proteinin kesilerek mitokondriyal yolağın tetiklenmesi vekaspaz-3’ün aktifleştirilmesi gerekir. Tip II hücrelerde Bcl-2 ve Bcl-XL proteinlerinin fazla ifade edilmesi kaspaz-8 ve kaspaz-3 aktivasyonunu vedolayısıyla programlı hücre ölümünü engellemektedir. Bu çalışma ile, bugünkü bilgiler ışığında programlı hücre ölümünün tarihçesi, morfolojisi vebiyokimyası hakkında Türkçe bir derleme ile Türk öğrenci ve araştırmacılara katkı sağlamak amaçlanmıştır.
Programmed Cell Death; A Turkish review of literature
Programmed cell death is an important and essential cellular event to eliminate unwanted cells during embriyogenesis, metamorphosis, endocrine-dependent tissue atrophy and normal tissue turnover. This is an energy-depended processes and succeeded by the cells that play an active role in theirown death. There are two major programmed cell death signaling pathways, the mitochondrial or intrinsic pathway and the death reseptor or extrinsicpathway. At the same time, perforin/granzyme dependent cell death can be included in the programmed cell death pathways. All the three pathways ofprogrammed cell death come together on the caspase-3/7 activation and on the events that occur after the programmed cell death such as chromatincondensation, DNA fragmentation degradation of nuclear as well as cytockeleton proteins etc. The cell death initiated by the external stimuli, theextrinsic pathway, occurs in two different way depend on the cell response to these stimuli. In this pathway, Type I called cells require activation ofcaspase-8 at the DISC (Death Inducing Signaling Complex) followed by activation of caspase-3. In type II cells, DISC formation is impaired andreduced active caspase-8 is generated. Active caspase-8 cleave and activate the cytosolic protein BID wich triggers mitochondrial pathway to generateactive caspase-3 to succeed programmed cell death. Overexpression of Bcl-2 and Bcl-XL blocked caspase-8 and caspase-3 activation as well asprogrammed cell death in Type II cells. In this review, we aim to provide a general overview of current knowledge on the process of programmed celldeath including history, morphology and biochmeistry as well as contribute by a Turkish review to the Turkish students and scientists.
