Ege bölgesi elit sporcularının ACTN3 R577X genotip dağılımının araştırılması

Amaç. Ege bölgesi elit sporcularının genetik alt yapılarının spordaki performansına olumlukatkısını moleküler yöntemlerle araştırmayı hedefledik. Yöntem. Moleküler genetik çalışma 105gönüllü elit sporcu ve 105 gönüllü kontrol (sedanter bireyler) grubuna uygulandı. Her birgönüllüden EDTA içeren tüplere 5‟er mL kan alındı. DNA izolasyonu, standart protokollere göreyapıldı. Polimorfizm, Polimeraz Zincir Reaksiyonu (PZR) ve Restriksiyon Uzunluk Polimorfizmi(Restriction Fragment Leight Polymorphism=RFLP) yöntemleri uygulanarak saptandı.Çalışmamızın istatistiksel analizleri SPSS 15,0 programı kullanılarak değerlendirildi. Bulgular.Elit sporcuların RR ve RX genotiplerinin dağılımları, sedanter bireylerden önemli ölçüde farklı(χ2= 17,9; p=0,000) bulundu. Sonuç. Ege Bölgesinde ACTN3 R577X genotip dağılımının, elitsporcu ve kontrol grubundaki prevalansının araştırılması Ege bölgesinde ilk defa yapılan birçalışmadır. Araştırmamızın ileride bu alanda yapılacak çalışmalara öncülük yapacağınıdüşünmekteyiz.

Investigation of ACTN3 R577X genotype distribution of elite athletes in the Aegean region

Aim. The aim of our study is to investigate whether the genetic base of elite athletes in Aegeanregion have positive effects on their performance in sports by molecular techniques. Methods.Molecular genetic study was applied to the 105 volunteer elite athletes and 105 volunteer controlgroup (sedantery individuals) individuals. Five milliliter blood samples were taken from eachvolunteer individuals into the tubes containing EDTA. DNA isolation was performed according tothe standard protocols. Polymorphism was determined by Polymerase Chain Reaction (PCR) andRestriction Length Polymorphism (RFLP) techniques. The statistical analysis of our study wasperformed by using SPSS 15.0 program. Results. Significant difference (x2 =17.9; p=0.000) wasfound in the distribution of RR and RX genotypes between the elite athletes and sedentaryindividuals (χ2= 17,9; p=0,000). Conclusion. This is the first study in the Aegean regioninvestigating the prevalence of ACTN3 R577X genotype in elite athletes and sedentaryindividuals. We think that our study will initiate further researches on this subject.
