Amaç: Çalışmamızda, farklı konsantrasyonlardaki imipenem ve meropenemin tek başlarına ve vankomisin ile kombinasyonlarının, standart suş Staphylococcus aureus ATCC 29213’e ve klinik örneklerden elde edilen metisiline dirençli Staphylococcus aureus (MRSA) ve metisiline duyarlı Staphylococcus aureus (MSSA) suşlarına karşı, postantibiyotik etkilerinin (PAE) araştırılması amaçlanmıştır. Yöntem: PAE tespitinde bakteriler, tek başına ve kombinasyon halinde antibiyotiklerle 1 saat temas ettirilmiş, sonra santrifüjleme ve yıkama yapılarak antibiyotik ortamdan uzaklaştırılmıştır. Daha sonra bakteriler katyon ayarlı Mueller Hinton Broth (KAMHB) besiyerine aktarılmış ve her saat başı Tryptic soy agar (TSA) besiyerine ekilmiş ve plak sayım yöntemiyle koloni sayımı yapılarak PAE süresi belirlenmiştir. Bulgular: İmipenem, meropenem ve bu antibiyotiklerin vankomisinle kombinasyonları Staphylococcus aureus ATCC 29213, MRSA ve MSSA suşlarında belirgin PAE süreleri göstermiştir. Sonuç: Farklı antibiyotik konsantrasyonlarında farklı PAE değerleri saptanmıştır.

Objective: In our study it was aimed to investigate the in vitro postantibiotic effect (PAE) of different concentrations of imipenem and meropenem, alone and in combinations with vancomycine against a standart strain of Staphyloccoccus aureus ATCC 29213 and clinically isolated methicillin susceptible Staphyloccoccus aureus (MSSA) and methicillin resistance Staphyloccoccus aureus (MRSA) strains. Method: During PAE experiment, the bacteria were exposed to antibiotics alone and in their combinations for one hour, then the antibiotic was removed from the culture medium by centrifugation and by washing the cells. After that, bacteria were transferred to Cation Adjusted Mueller Hinton Broth (CAMHB) medium and then bacteria were spread on Trypticase soy agar (TSA) medium for intervals of one hour. The PAE was determined by viable plate count method. Results: Imipenem alone, meropenem alone and the combination of two antibiotics with vancomycine against Staphyloccoccus aureus ATCC 29213 strain, MRSA and MSSA showed a significant effect on PAE. Conclusion: Consequently at different antibiotic concentrations, PAE was found to be different.
