ÖZET Amaç: İnfertil bireylere ait sperm hacim/uzunluk oranları ve spermiyogenezin fizyolojik sürecinde aktin bağlayıcı protein olan ve bir çapraz bağlayıcı olarak kortikal hücre iskeleti ile plazma membran proteinleri arasında bulunan ezrin, ile bir transmembran glikoproteinlerin olan N-kaderin kolakizasyonlarının araştırılması gerçekleştirilmiştir. Gereç ve Yöntemler: N= 18 infertil bireyde hacim/uzunluk oranları ve sperm sayımı gerçekleştirilen semen önekleri sperm yayma preparatları Kruger Kriterlerine göre mikroskop altında incelendi. Her grup için sperm uzunluk ölçümleri gerçekleştirildi, immünofloresan boyama ile ezrin ve N-kaderin lokalizasyon ve ekspresyonlarıdeğerlendirildi. Bulgular: Sperm uzunluklarının hacim yoğunluklarına oranla azaldığı ve sperm uzunluğunun sperm sayısı ile arasında anlamlı korelasyon olduğu tespit edildi. Sperm kuyruk uzunluğu anomalisinin infertilite üzerinde etkin bir durum olduğu ancak ezrin ve N-kaderin lokalizasyon ve ekspresyonlarında control grubuna göre anlamlı fark olmadığı tespit edildi. Sonuç: Sperm sayısı/hacim ve sperm uzunluk oranlarının infertil bireylerde spermiyogenezin fizyolojik sürecine bağlı olarak gelişebileceği ilk kez bu proje ile gösterilmiş olup, sperm morfolojik özelliklerinin semen hacmi ile ilişkili olabileceği ve infertil bireylerde belirli fonksiyonel proteinlerin tespitinin gerçekleşmesine yönelik çalışmaların gerekliliği vurgulanmıştır. Anahtar Kelimeler: Erkek infertilitesi; Semen analizi; Sperm uzunluğu; Ezrin, N-kaderin ABSTRACT Aim: The aim of this study was to investigate the sperm volume/lenght ratios of infertility individuals and N-cadherin co-localizations, which are the actin binding protein in the process of spermiogenesis, and ezrin, which are between the cortical cell skeleton and plasma membrane proteins, as a cross-linker. Material- Method: Semen samples taken from infertilite individuals (N=18) examined by microscopy after staining and fixation. The volume/length ratio was performed. Sperm morphology was examined as sperm smear preparations according to Kruger Criteria. Sperm length measurements were performed for localization and expression parameters of ezrin and N-cadherin were examined by immunofluorescence staining. Results: The sperm length of the patients with similar morphological criteria decreased with respect to volume and there was a significant correlation between sperm length and sperm count. It was found that sperm tail length anomaly was effective on infertility, but there was no significant difference in localization and expression of ezrin and N-cadherin compared to control group. Conclusion: It has been shown for the first time that sperm count/volume and sperm length ratios may develop depending on the process of spermiogenesis in infertile individuals, it is emphasized that sperm morphological features may be related to semen volumes and necessity of studies fort he detection of certain functional proteins in infertile individuals is emphasized.
