IBA ve bakteri (Agrobacterium rubi) uygulamalarının Kütahya vişne çeşiti çeliklerinin köklenmesi üzerine etkisi
1999-2000 yılları arasında yürütülen bu araştırmada, materyal olarak Kütahya vişne çeşidine ait odunsu "çelikler kullanılmıştır. Çelikler dinlenmeye giriş döneminde (25 Ekim) alınmıştır. Alınan çeliklere yalnız ve kombinasyon şeklinde 2000, 4000, 6000 ppm IBA ile Agrobacterium rubi'nin 3 farklı ırkı (Al, A16, A18) uygulanmıştır. IBA solüsyon halinde hızlı daldırma yöntemiyle, Agrobacterium rubi ise yine solüsyon halinde $1x10^8$ bakteri/ml konsantrasyonunda hazırlanarak çeliklere uygulanmıştır. Uygulama yapılan çelikler serada ınisfleme sisteminde, içinde perlit bulunan ortama yerleştirilmiştir. Yaklaşık 3 ay süre ile mistleme ortamında bırakılan çelikler bu süre-nin sonunda sökülmüş ve köklenme oranlan (%), kallus oluşturma oranları (%), 1. sınıf kök oranlan ve çelik başına kök sayısı (adet) belirlenmiştir. Araştırma sonunda kontrol uygulamasında köklenme elde edilemezken (%0), diğer uygulamalarda farklı oranlarda köklenme elde edilmiştir. Yapılan uygulamalar i-çerisinde en yüksek köklenme oranı 2000 ppm IBA+A16 uygulamasından (%70.0) el-de edilmiştir.
Effect of IBA and bacteria (Agrobacterium rubi) on the rooting of cuttings of sour cherry cv. Kütahya
This research was carried out between 1999-2000 using stem cuttings of sour cherry cv. Kütahya as the material. The cuttings were taken in the beginning of dormancy period (October 25th), and treated with IBA only at different concentrations (2000, 4000 and 6000 ppm) and in combinations of three Agrobacterium rubi strains (Al, A16, A18). Quick dip method was used for application of IBA solution. Agrobacterium rubi strains were applied in the form of bacterial suspension prepared in sterile water at the concentration of Ixl08.bacteria/ml. The treated cuttings were placed in the mist propagation system including perlitte medium. After three months the cuttings were uprooted and evaluated for rooting percentage (%), callusing (%), 1st class root ratio (%) and number of roots per cutting. No rooting was observed on the cuttings of sour cherry with the control treatment whereas the average percentage of rooting on the cuttings of other treatments was different. The highest rooting percentage was determined as 70%, obtained from the application of IBA (2000ppm)+A16.
