Sıçan akciğeri ve böbreği mikrozomlarında ilaç metabolize eden enzimlerin aktiviteleri üzerine çalışmalar

Bu çalışmada sıçan akciğer ve böbreği mikrozomal fraksiyonlarında sitokrom P450 'ye bağımlı enzim sistemi üyelerinden 7-etoksiresorufin-O-etilaz (EROD), 7-pentoksiresorufin-O-etilaz (PROD), eritromisin-N-demetilaz (ERND), kumarin 7-hidroksilaz (Coh) ve p-nitrofenol hidroksilaz (PNP) enzim aktiviteleri için optimum pH, protein miktarı, substrat konsantrasyonu ve inkübasyon süresi saptandı. Sıçan akciğer ve böbrek mikrozomlarmda bu enzim aktiviteleri için farklı reaksiyon koşullarına gereksinim olduğu saptandı.

Studies on the activities of drug metabolizing enzymes in lung and kidney microsomes of rat

The optimum conditions (pH, protein amount, subtrate concentration and incubation time) of 7-ethoxyresorufm-O-deethylase(EROD), 7-pentoxyesorufin-O-deethylase(PROD), eryhtromycin-N-demethylase (ERND), coumarin 7-hydroxylase (Coh) and p-nitrophenol (PNP) activities in the lung and kidney of rat were determined in microsomes. In this study, it was found out that, different reaction conditions are required for these enzyme activities in rat hind and kidney microsomes.
