Cynara syriaca'dan elde edilen flavonoidlerin lösemi hücreleri üzerine etkisi

Flavonoidler doğada birçok bitkide, yaygın olarak bulunan düşük molekül ağırlıklı, bifenolik bileşiklerdir. Bu gruba dahil olan glikozidler kardiyovasküler sistem düzenleyicileri olarak çok eskiden beri kullanılmaktadır. Son yıllarda ise antienflamatuar etkileri, serbest radikal temizleyici özellikleri ve prokarsinogenezde inhibitör etkileri araştırılmaktadır. Bu çalışmada Cynara syriaca'dan elde edilen kafeik asit, apigenin, apigenin-7-glikozid, luteolin, luteolin-7-glikozid ve sinarinin akut lenfoblastik lösemili hastalardan alınan blastik hücreler ile lösemik hücre dizileri (DG75, B95, BB58, K562) üzerindeki etkileri araştırıldı. Altı ayrı dozda (500, 50, 5, l, 0.05, 0.005 mg /ml ) flavonoid belli sayıdaki hücreler ile in vitro kültür yapılarak, 3 gün sonunda canlı hücreler MTT ( methyl-thiazol-tetrazolium-5 mg/ml) ile bayandı. Forınazan kristalleri DMSO ile çözülüp spektrofotometrede ölçüldü. Kontrole göre proliferasyon ve sitotoksite indeksleri hesaplanarak, flavonoidlerin etkisi test edildi. İstatistiksel analizler için Student-t testi kullanıldı. 500mg/ml kafeik asit hem hasta lösemi hücrelerinde (23,5+/- 12,3) hem de standardize edilmiş lösemi hücre serilerinde (45 +/-30,4) proliferatif etki gösterdi. Sinarin (20,45+/-12,3), apigenin (19,5+/-9,6), luteolin-7-glikozid (19,5+/-12,1) ve apigenin-7-glikozid ise (20,9+/-8,6) oranında lösemi hastalarında sitotoksik etki gösterdi.

Effects of flavonoids obtained from Cynara syriaca on leukemic cells

Flavonoids are bifenolik compounds with low molecule weight, which are found most plants in nature. Glycosides which of their subgroubs are used as regulators of cardiovascular sistems for a long time in past. Recently, flavonoids are studied anti-inflammatory effects, free radical scavenging activities and inhibitor effects in procarsinogenesis. This study is investigated effects of caffeic acid, apigenin, apigenin-7-glikozid, luteolin, luteolin-7-glikozid, cynarin, which are obtained from cynara syriaca, on leukemic cell line (K562,DG75,BB58,B95) and blasts of patients with acute lymphoblastic leukemia. Leukemic cells are incubated with six different doses of flavonoids (500, 50, 5, 1, 0.05, 0.005 mg /ml) for 3 days, Later cultures treated with MTT that viable cells are dying After formazan crystals in cells are solved with DMSO, are evaluated in ELISA spectrophotometer. According to control, proliferation and cytotoxicity index are accounted. The test of Student-t is used for statistical analysis effects of flavonoids. 500mgml caffeic acid show proliferative effects in both leukemic cells (23,5+/- 12,3) and cell lines (45+/-30,4). Cynarin (20,45+/-12,3), apigenin (19,5+/ -9,6), luteolin-7-glycosid (19,5+/-12,1) and apigenin-7-glycosid (20,9+/-8,6) suggest cytotoxic effect in patients with acute lymphoblastic leukemia.
